Excited to share a key part of my postdoctoral work in the Cravatt lab at @ScrippsResearch, published today in #NatureChemistry! Huge thanks to collaborators #brumelillo, #RachelHayward, #VividionRx, and all coauthors. Special appreciation to #HHMINEWS Hanna Gray Fellows Program, #CancerGrand and #theNCI for their financial support. Our study addresses the gaps in residue-directed ABPP platforms by integrating a novel protein-directed ABPP approach. This allowed us to identify over 300 stereoselective ligand-protein interactions from our focused library of tryptoline acrylamide stereoprobes. We characterized ligands for a diverse class of proteins, including those that (1) disrupt MAD2L1BP interactions and delay mitotic exit— an adaptor protein essential in cancer, and (2) target a non-catalytic cysteine in the serine hydrolase, AFMID, a key enzyme in tryptophan metabolism. This work further highlights how covalent chemistry and stereochemical controls can reveal the functional outcomes of newly discovered ligand pockets, even with moderately potent probes. 🥇 Last week, I won a GOLD medal in a Kaggle competition of Leash Bio, placing 8th out of around 2000 teams! This was my second competition in the field of life sciences, where I earned a GOLD. And now I have become a Kaggle Master! The problem was to predict which drug-like small molecules (chemicals) will bind to three possible protein targets, which in turn may help develop a drug against blood pressure, diabetes progression, and cancer. There were 133 million molecules being tested in the wet lab that participants used for training and evaluation of the ML models. This is one of the proofs, among a few others, that we had in our company showing that the similarity of molecules is an important factor that should be considered for preparing the benchmarks in at least the tasks of small molecule properties prediction.
