Kicking KRAS to the kerb It's not so long ago when we saw numerous companies vying to to drug targets such as VEGF and EGFR – with limited success in many cases.  Even today there are not too many approved drugs in these two categories relative to the number of agents which ended up in 'dog drug heaven'.  The dreaded phrase was originally coined by Dr Pat LoRosso while at Karmanos Cancer Centre in Detroit; she's now at Yale and the incoming AACR President.  Befitting the lady's style – she's one of the nicest and kindest oncologists I've ever met – it seems a much more pleasant way to describe abandoned or discontinued anticancer agents than referring to the veritable dustbin of failed dreams. The latest posse of drugs needing some stringent culling or shakeout will likely come from the KRAS niche because the market is unlikely to be forgiving commercially of any drug not moving the needle in a substantial fashion.  The good news is there are some gems in the rough for those who care to look more carefully and closely at what's needed to move forward.  One of them is the focus of our latest company interview from AACR – what are Frontier Medicines doing differently and why does it matter? The U.S. Food and Drug Administration (FDA) is conducting an investigation into certain cancer therapies involving chimeric antigen receptor T-cell (CAR-T) therapies, following reports of serious safety concerns. This probe specifically targets treatments developed by companies including Gilead Sciences, Johnson & Johnson, and Novartis. The safety issues in question involve reports of T-cell malignancies, encompassing various blood disorders like lymphomas and leukemias, after patients received CAR-T treatments. Since 2017, six CAR-T cell therapies have been FDA-approved, all intended for treating blood cancers such as lymphomas and certain types of leukemia. These therapies include Bristol Myers Squibb's Breyanzi and Abecma (in partnership with 2seventy bio), J&J's Carvykti (developed with Legend Biotech), Novartis' Kymriah, and Gilead's Tecartus and Yescarta. The safety concerns raised could potentially impact these treatments future.

Our latest research, driven by PandaOmics, identified MYT1 as a promising therapeutic target for breast and gynecological cancers, with our lead compound 21, demonstrating remarkable selectivity and promising in vivo antitumor efficacy. Through structure-based drug design, we optimized potency and physical properties, offering a potential breakthrough in innovative treatment options for these deadly diseases. ST-001 has received Orphan Drug Designation from the FDA for the treatment of CTCL (a form of T-cell NHL). This designation is given to drugs targeting rare diseases affecting fewer than 200,000 people in the US. Orphan Drug Designation provides incentives to develop treatments for these conditions, which often lack adequate therapeutic options.
