In my previous post, I mentioned the numerous assays that different labs use for early developability assessments. At PAIA Biotech, we’ve decided to incorporate as many of these assays as possible into our technology to offer a flexible toolbox, allowing each customer to select their preferred options. Here are our current standard tools: Hydrophobic Interaction Assay (HIC): The crowd favorite 🏆 – why? 1️⃣ Hydrophobicity data is essential for everyone. 2️⃣ This assay correlates well with HPLC and is simple to evaluate. 3️⃣ It’s 100x faster than chromatography. Ovalbumin Binding Assay (OVA): Popular for polyspecificity – why? 1️⃣ The Tessier lab found Ovalbumin to be better suited than other reagents. 2️⃣ We confirmed that OVA binding correlates closely with PSR. 3️⃣ OVA is a well-defined reagent, which makes it ideal for robust assays. Heparin Binding Assay (HEP): Growing in popularity 🚀– why? 1️⃣ Papers by Roche and recently Amgen highlighted its usefulness. 2️⃣ The assay correlates well with chromatography and is easy to evaluate. 3️⃣ Huge increase in throughput compared to chromatography. Cation Exchange Assay (CEX): Still in its early stages – why? 1️⃣ Few labs routinely run CEX chromatography at this point. 2️⃣ Some may believe that pI provides similar information, but we wonder if this assay offers a more realistic view of how a molecule interacts with a chemically defined negative surface. 🤔 Would be great to get some more insight into this! Next week, I’ll cover the less frequently requested tools in our toolbox—but who knows, there might be a hidden gem among them!
