From the abstract of this paper: Here, we develop a localized cytokine therapy that elicits profound anti-tumor immunity by engineered targeting to the ubiquitous leukocyte receptor CD45. We designed CD45-targeted immunocytokines (αCD45-Cyt) that, upon injection, decorated the surface of leukocytes in the tumor and tumor-draining lymph node (TDLN) without systemic exposure. αCD45-Cyt therapy eradicated both directly treated tumors and untreated distal lesions in multiple syngeneic mouse tumor models. Mechanistically, αCD45-Cyt triggered prolonged pSTAT signaling and reprogrammed tumor-specific CD8+ T cells in the TDLN to exhibit an anti-viral transcriptional signature. CD45 anchoring represents a broad platform for protein retention by host immune cells for use in immunotherapy.

🗞️Have a look at our latest case study in which we helped Medicines for Malaria Venture (MMV) generate an antimalarial with reduced toxicity in less than an hour. 🌩️MMVs lead candidate in their aryl piperazine series exhibited high CYP450 activity which may cause adverse interactions with other drugs in humans. 🌤️Using DeepMirror, we generated 1,000+ compounds and quickly identified a compound with high antimalarial activity but lower CYP450 activity than the lead. This compound was subsequently validated experimentally by MMV testing entities. 🤷🏻♀️Why does this matter? With artificial intelligence reads of chest X-rays and an affordable TB test to confirm the diagnosis, mass screening and testing will be feasible and affordable in many countries.

Even slight variations in instrument settings, sample handling, or reagent quality can influence the results of a cytometry run. That’s why it’s crucial to perform intra-run precision tests to ensure these factors remain stable and the data generated is reliable and reproducible. Using our 25-marker spectral flow cytometry assay, we measured the frequency of immune cell populations across three samples of peripheral blood mononuclear cells from each of three healthy donors in a single run. Similar to our inter-run results, we achieved an average coefficient of variation (CV) of just 3.6%, surpassing the industry standard of 20%.
