Urine analysis of a patient!! This transparent liquid that we excrete contains thousands of bacterias, pus cells , dead RBCs and epithelial cells. In rare cases it might also carry some parasite ( sometimes moving) 😱. Urine analysis is done by microscopic viewing of urine pellet that we receive after centrifugation. 🔬 When viewed, I ovserved Bacteria cells Epithelial cells Waxy casts fungi RBCs in clumps calcium crystals etc. Its like a new adventure, working with different samples each day and preparing reports. 🌱 Exciting Project Overview: Advancing Microbiome Research with PacBio Sequencing 🌱 I’m thrilled to share a project led by an AGRF intern Ynna Hernandez-Europa from QUT (Queensland University of Technology) that explores the vital role of microbiomes - communities of microorganisms like bacteria, fungi, and viruses that influence our health, agriculture, and the environment. Imbalances in these microbial communities can be linked to various diseases and significantly impact agricultural productivity and soil health. "My PhD aims to understand the biology of an insect pest (Heliococcus summervillei) that is decimating pastures across QLD and NSW. This AGRF internship is an excellent opportunity to collaborate with experts in the field of sequencing and explore how current technologies could benefit my research. I'm keen to dive into long-read sequencing and analysis - having only worked on Sanger and short-reads - and I'm ready to tackle the challenge of troubleshooting code in a fast-paced, real-world setting!" Ynna explains. This project leverages PacBio long-read sequencing technology to profile microbial communities by targeting the bacterial 16S rRNA gene. Traditional short-read sequencing often struggles to accurately classify complex microbial environments, while PacBio’s long-read capabilities provide full-length 16S sequences, improving our ability to identify microorganisms down to the species level. While the PacBio platform offers incredible potential, it also presents bioinformatic challenges such as error correction, taxonomic assignment, and data processing speed. Our aim is to refine and optimise bioinformatic workflows to effectively handle PacBio 16S data, leading to more accurate and efficient microbial profiling. Expected Outcomes: 🔹 Insights into the benefits of PacBio long-read sequencing for microbiome studies. 🔹Established protocols for future microbial profiling projects. 🔹Enhanced accuracy in detecting and classifying microbial species. This internship based in our Brisbane lab offers a fantastic opportunity to gain hands-on experience in cutting-edge genomic technologies and bioinformatics analysis.
