I'm pleased to see the updated USG Dual Use Research of Concern (DURC) policy better clarifies review and oversight of pathogen research. I found it interesting that the 2024 version seems to take a stronger stance on research institutions that do not receive federal funding compared to the 2015 policy: 2015: "Institutions that do not receive USG funds for life sciences research... are not subject to oversight as articulated in this Policy; however, they are strongly encouraged to implement internal oversight procedures consistent with the culture of shared responsibility underpinning this Policy." https://lnkd.in/gF7hXNxt 2024: "Research institutions that do not receive any federal funds for life sciences research... are strongly encouraged to implement oversight procedures consistent with the culture of shared responsibility underpinning this Policy. What we can't ignore is that the way the U.S. handled this Covid pandemic was itself a failure to learn. If we use this systems thinking frame of reference, what is the purpose of a system that demonstrates repeated & potentially catastrophic "failures" relating to pandemics? Out of 27 'breakthroughs' reported in the UK national press, only 1 resulted in human benefit. A study by Cruelty Free International investigated whether highly publicised animal-based experiments had led to any new treatments, 25 years on. The results are clear. 💬 "Overspeculation and exaggeration of human relevance was evident in all the articles examined. Of 27 unique published ‘breakthroughs’, only one had clearly resulted in human benefit. 20 were classified as failures, 3 were inconclusive and 3 were partially successful." PBPK Modeling Demonstrates That a Time Exposure Adjustment is Unnecessary for an Acute Manganese Health Guideline For the purpose of resolving uncertainty in determining an acute manganese inhalation toxicity criterion, ToxStrategies’ Camarie Perry, Ann Holbrow Verwiel, Tammie Covington, and Deborah Proctor used the most current published PBPK model for manganese to confirm that a time-adjustment factor is unnecessary when extrapolating animal bioassay exposure conditions for assessing acute (24-hour) human exposure, including among potentially exposed children. This recently published work (https://lnkd.in/gmKxQHnJ) expands upon a previous publication (https://lnkd.in/g39G6KDS) by ToxStrategies scientists and demonstrates that a proposed 4-fold time adjustment factor is not necessary to ensure protection of public health.

How safe is PFOA for humans? A recent study attempted to estimate the dose of PFOA that does not pose a risk to human health. This challenging goal is complicated by the large number of epidemiological studies with highly variable results, as the authors point out. A team of 24 scientists from eight countries reviewed the relevant data and independently developed ranges of estimated safe doses of PFOA. The scientists concluded that the available epidemiological information is insufficient to provide a reliable basis for assessing a safe human dose of PFOA. Using dose-response data from five studies of laboratory animals exposed to PFOA, the researchers estimated that PFOA doses between 10 and 70 ng/kg per day are not hazardous to human health. https://lnkd.in/d9rkAbcB While I agree with the researchers that deriving safety estimates from epidemiological studies is problematic, I disagree with their conclusions based solely on animal studies. In fact, PFOA bioaccumulates in the human body and persists much longer than it does in laboratory animals.
