People interested in filtration: let’s talk flow rates! Downstream processing is consistently referred to as a major bottleneck of bioprocessing workflows, especially when it comes to primary and secondary clarification of a culture or fermentation. Depth filters might work well at the start of a run, but flow rates will inevitably drop as time goes on and fouling becomes a reality. If you need to replace the spent depth filters with fresh new ones, you’re not only increasing the run time but also compromising sterility of the process. Quick processing times are not exactly a selling point of centrifuges either, as increasing their flow rate almost always comes at the expense to the filtrate clarity. With the FUNDALOOP® , we have processed mammalian culture and microbial fermentations between 2-3x faster than depth filters. Ask me for the data! How is this possible? Our proprietary filter cloth and backflush step. We have a cloth regeneration step built into our automated system, so the filter cloth can remain good as new from start to finish. Once solids begin to build up on the cloth and significantly reduce the flow rate, WFI or buffer is injected in the reverse direction of the cloth to dislodge the filter cake. The solids are then discharged as waste, and the resulting clean filter cloth is ready to start a new cycle at full capacity. This is what we call cyclical cake filtration, and it is a game-changer when it comes to process speed. Flow rates that are 2-3x faster than traditional methods is a serious incentive, but does our increased speed come at the cost of filtrate quality…? I’ll touch on this next week!
