It's #FluorescenceFriday and today we celebrate another milestone paper in the history of #microscopy: The first article that described #SingleMoleculeFRET (#smFRET) was published on the 25th of June 1996 in the journal #PNAS. #FRET, Fluorescence Resonance Energy Transfer, is a method where a fluorophore is excited by light at a certain wavelength, and it then immediately transfers energy to another fluorophore, which then emits energy in a different wavelength. FRET can only work if the two fluorophores are found in very close proximity, and is therefore useful in experiments where scientists want to measure molecular distances of bio-molecules. For examples, researchers can label two proteins and then test whether they bind each other or not, based on the FRET signal. Single molecule FRET is a #SuperResolution approach which relies on FRET signals of single fluorophores, and enables scientists to study the dynamics of biological interactions, including in living organisms. To quote the historic paper from the group of Prof shimon weiss, formerly from the Berkeley Lab, who authored the paper: "Conventional (i.e., ensemble) FRET measurements, which rely on signal averaging over many molecules, cannot detect dynamic events such as relative motion between donor and acceptor molecules when these event are not synchronized between molecules, as is often the case. The ability to measure FRET on a single pair potentially allows the study of time-dependent phenomena such as protein and molecular motor conformational changes."
