Preparation Techniques of Polypeptide - Genetic Engineering Recombination. The technology for producing #peptides by this method has become mature and has the advantages of not being limited by production raw materials and strong expression directionality. Gene recombination methods are often used to synthesize peptides with relatively long peptide chains, especially peptides composed of hundreds of amino acids. The #genetic recombination method can recombine the gene sequence that regulates polypeptide synthesis into the genetic material of microorganisms, etc., and translate and express the genetic material in the recipient organism. The yield and purity of the polypeptides obtained by this method are greatly improved, and it is currently one of the effective ways to obtain certain specific biologically active peptides in large quantities. However, it is limited to the synthesis of macromolecular peptides, and it is difficult to express short peptide chains. Recombinant protein expression system - yeast expression system. According to the type of host cell, gene expression systems can be divided into prokaryotic expression systems, yeast expression systems, mammalian expression systems, and insect expression systems. Yeast is a eukaryotic microorganism whose cells have the ability to express foreign proteins and can reach a very high cell density state. Expressing foreign proteins in yeast can undergo post-translational modifications, such as phosphorylation and glycosylation. Since yeast host cells combine the excellent properties of bacterial and higher-order organism host cell expression systems, they are a good choice when foreign proteins cannot be successfully expressed in E. coli. Yeast strains used for foreign protein expression include Saccharomyces cerevisiae, Schizosaccharomyces pombe, Pichia pastoris, and Kluyveromyces lactis.
