Developmental and reproductive toxicity (DART) is an area of #toxicology that is required to assess the impact of new chemicals on adult fertility and embryo development. Industries rely on DART assessments to safeguard human health, adhering to regulatory standards like those outlined in the #ICHS5 guideline. However, the current regulatory landscape for DART assessments is not without its challenges. Traditional methods rely heavily on animal testing, which comes with significant ethical concerns, high costs, and limited insights into the underlying mechanisms of toxicity. 🧬 📝 Our latest #research details an alternative approach to animal testing, which can be used to screen for DART hazards at the early stages of compound development. Check out our latest article on a mechanism-based assessment of endocrine disrupting properties of #PFOS, based on AOPs and NAMs! 🎉 This project was A LOT of work and we identified several aspects that are important for the development of Next Generation Risk Assessment (#NGRA) of endocrine disruptors (EDs). In this paper, we compare a standard ED assessment against a mechanism-based assessment that did not utilize any in vivo data. Both assessments investigate thyroid disruption and developmental neurotoxicity caused by PFOS exposure. We utilize a thyroid-relevant Adverse Outcome Pathway (AOP) network to integrate data from New Approach Methodologies (NAMs) and to postulate a Mode-of-Action. In a significant proportion of the cases, the 2nd species in not needed. And in some rare case, the toxic effects can be seen only in this second species. Understanding species-specues mechanisms of action or effects of matériel toxicity is of most importance to study toxicity and prevent the toxic effectsnin humans. Our group recently published a peer-reviewed paper titled "A review of the mesotheliogenic potency of cleavage fragments found in talc" in the journal Toxicology and Industrial Health. In the paper, we reviewed the current state of the science regarding the factors influencing mesotheliogenic potency of cleavage fragments, with emphasis on those that may co-occur in talc deposits, including dimensional and structural characteristics, animal toxicology, and the most well-studied cohort exposed to talc-associated cleavage fragments.
