Surfactants are the most common additives in dissolution media. They can act as wetting agents or increase solubility, but their use isn't without challenges. Using too much surfactant can lead to in-discriminatory profiles, and even when using an appropriate amount you can have other technical challenges. Surfactants are helpful when dealing with poorly water soluble drugs. They increase wetting of hydrophobic drugs by lowering interfacial tensions between the solid drug particles and the dissolution media. At higher amounts above the CMC (critical micelle concentration), they increase the solubility of the drug itself. Since surfactants can alter the #solubility of the drug itself, you need to be careful not to add too much to the media and lead to artificially high release rates or indiscriminate dissolution profiles.There are many types of surfactants which are used in dissolution and have different activities based on their chemistry. They can be positively or neagatively charged, as well as neutral. When choosing a surfactant, as well as an amount to use, you should test multiple surfactant types in your dissolution media. Determine which surfactants, and at which levels, are able to help you achieve sink conditions of your drug. You can do this easily by weighing your label claim equivalent dose into a volumetric flask about 1/3 the volume of your dissolution media, filling it with your dissolution media containing #surfactant, and shaking it for an hour. You can create multiple #pH media, with multiple surfactants and create a very quick visual screening process for some potential media and surfactant combinations. If you find you have dissolved the drug, you can do a second wave of experiments backing down the amount of surfactant. Some common surfactants to try are SLS, CTAB, Tween, Triton X, and bile salts. Of these, #SLS is the most common - but not without issues.
