Interested in  cell dynamics in space and time? Check out this detailed protocol and guidelines for ultra-low-input spatial tissue proteomics from Fabian Cosca. The presented workflow allows the proteomic analysis of ultra-low-input archival material, with up to 2,000 quantified proteins from single hepatocyte contours and nearly 5,000 proteins from 50-cell regions in murine liver. Applied to human tonsil, the approach profiles various microregions, quantifies cell-type-specific markers, cytokines, and transcription factors, and offers insights into proteome dynamics making it valuable for biomedicine applications such as early disease profiling and drug target and biomarker discovery. #singlecellproteomics #proteomics #massspectrometry #tissue #FFPE #lasermicrodissection #spatialproteomics
