#DYK that #OnThisDay in 1926 Thomas Dale Brock was born, an American microbiologist who found the bacteria, Thermus aquaticus, which thrives at temperatures of 70°C (158°F) or higher, because all their enzymes are stable at very high temperatures. In the 1980s, biochemist Kary B. Mullis used this species of bacteria to help create the polymerase chain reaction (PCR), which requires cycles of extreme heating and cooling. PCR technology is now widely used to rapidly multiply traces of DNA millions of times to produce enough for analytical study. https://lnkd.in/dchnMJK #Microbiology #Biochemistry #ScienceHistory #PCRTechnology #DNAAnalysis #Microbiologist #Biochemist #ScientificDiscoveries #BacterialResearch

Storing Cells in Liquid Nitrogen "The Cryogenic Vault: Safeguarding Cell Lines in Liquid Nitrogen Liquid nitrogen is the gold standard for long-term cell storage. Here's why: Ultra-low temperatures: Inhibit cellular activity and prevent degradation. Vapor phase storage: Minimizes temperature fluctuations. Proper vial labeling: Essential for organization and traceability. Regular monitoring: Ensures optimal storage conditions. By following these guidelines, scientists can maintain the integrity of their cell lines for years to come." #cellculture #biotechnology #lablife #science #research #cellbiology #liquidnitrogen #storage #cryopreservation Cell Thawing Protocol "Bringing Cells Back to Life: The Art of Thawing Thawing cells requires precision and speed. Here's a basic protocol: Rapid thawing: Immerse the frozen vial in a 37°C water bath. Transfer: Quickly transfer cells to pre-warmed growth medium. Centrifugation: Remove harmful substances. Resuspension: Gently resuspend cells in fresh medium. Viability check: Assess cell health before further use. Remember, specific cell types may have variations in this process. Always follow established protocols." #cellculture #biotechnology #lablife #science #research #cellbiology #thawing #cryopreservation #protocol

Stopped-flow measurement of CO2 hydration activity by catalytic amyloids Liam R. Marshall, Olga Makhlynets Methods of Enzymology, 21 March 2024 https://lnkd.in/gfvAFqKe Abstract: With the ever-increasing rates of catalysis shown by catalytic amyloids, the use of faster characterization techniques is required for proper kinetic studies. The same is true for inherently fast chemical reactions. Carbon dioxide hydration is of significant interest to the field of enzyme design, given both carbonic anhydrases’ status as a “perfect enzyme” and the central role carbonic anhydrase plays in the respiration and existence of all carbon-based life. Carbon dioxide is an underexplored hydrolysis substrate within the literature, and a lack of a direct spectroscopic marker for reaction monitoring can make studies more complex and require specialist equipment. Within this article we present a method for measuring the carbon dioxide hydration activity of amyloid fibrils. Acknowledgments: This work was supported by The National Institutes of Health.
