We are pleased to announce the presentation of a “Trial-in-Progress” (TiP) poster from our Phase 1/2a trial of VB10.16 C-03 in HNSCC at ESMO - European Society for Medical Oncology 2024.   The open-label, dose-finding trial evaluates the safety, immunogenicity, and anti-tumor activity of VB10.16 in combination with MSD’s PD-1 inhibitor KEYTRUDA® (pembrolizumab) in patients with HPV16-positive, PD-L1-positive, recurrent or metastatic head and neck squamous cell carcinoma (HNSCC) eligible for pembrolizumab monotherapy. The trial consists of two consecutive phases: a dose escalation phase (Phase 1) and a dose expansion phase (Phase 2a). #ResearchUpdate #InnovationInProgress #HOPEforPACS2syndrome Introduced in 2013 by Anne Carpenter and her team at the Broad Institute of MIT and Harvard, cell painting is a method providing unbiased insights into the potential therapeutic effects of a tested compound, particularly for disorders with limited basic science coverage (= you know close to nothing about the disease biology, as we did about PACS2 syndrome). We initiated cell painting drug screening a few months ago, led by Charles River Laboratories. Fortunately, we encountered no major obstacles along the way - a relatively uncommon situation in scientific endeavors 😉. In October, we even had the pleasure of consulting the initial results with Anne Carpenter. We have just completed the primary screen, and the results have given us a strong sense of optimism regarding the future of drug repurposing for PACS2 syndrome 😊. In fact, we are optimistic enough to commence preparations for clinical trials in 2025. -- If you want to contribute to the potential breakthrough for PACS2 community, support us: https://lnkd.in/dMAguf38 -- FACTS Charles River Laboratories successfully screened two extensive libraries comprising over 5000 compounds being registered drugs or in clinical trials. Leveraging the power of cell painting, they've narrowed down this vast pool to 70+ compounds — our promising "hits" capable of restoring diseased cell morphology by at least 40%. What's even more thrilling is that approximately 20 of these compounds align with a solid biological perspective. Some also hold the potential to either provide deeper insights into the disease mechanism of action or lead us to new pathways. Of course, there will be a few dead ends in the mix (#science), but these are the results we dared to dream of when embarking on this screening journey. To say the team is excited would be an understatement 😊   WHAT NOW Further pre-clinical tests: dosage response (vs toxicity) and drug sequencing for the start. In parallel, our Partner, Perlara PBC, is set to conduct a thorough analysis of the biological significance of the detected hits. Cereno Scientific’s innovative study design of the Phase II study of CS1 in PAH published in the renowned medical journal Pulmonary Circulation.   The research article describes how the design of clinical trials is important to fully understand the mechanisms and benefits of new treatments. Numerous potential pharmacological treatments for PAH have shown promise in preclinical models but have failed to show clinical benefit in humans or have been associated with poor safety and tolerability. A novel feature of the CS1 Phase II study is that the patients are monitored for pulmonary hemodynamics and right heart ventricle function on a daily basis with the implanted CardioMEMS HF technology. Furthermore, in the study, which has safety and tolerability as primary objective, a wide array of functional, hemodynamic, and structural measures and biomarkers as well as patient-reported outcomes are being collected to provide an insight into not only the treatment effects but the disease progression itself. The research article concludes that CS1 represents a potential novel disease-modifying treatment for PAH.   Read the full press release via link.

Interesting new piece on #BioelectronicMedicine in MIT Review by Sally Adee. It begs the question I’ve been thinking about a lot: Are we just sending signals through the wires of the body, nerves, for our therapeutic effects and limiting side effects, or fundamentally changing the wires (or most likely both)?
