On March 21st, 2024, during the Open Science Bar meeting titled "Digital Spatial Transcriptomics as a Powerful Tool for Clinical Cancer Research," Dr. Silvia Brich and Dr. Katia Todoerti presented a technical and analytical overview, highlighting the advantages and limitations of the GeoMx-Digital Spatial Profiler (DSP). They offered crucial insights into experimental design considerations for translational oncology research utilizing this tissue-based high-plex profiling approach. Recent studies have demonstrated the efficacy of GeoMx in identifying biomarkers across various tumor types, treatment contexts (such as immunotherapy), and tissue compartments. Scientists at St. Jude Children's Research Hospital - ALSAC have identified 156 potential CAR-T cell therapy targets in brain and solid tumors – providing new possible targets for cancer treatment. The comprehensive analysis included 1,532 pediatric tumor samples and 7,460 normal tissue samples, and revealed cancer-specific genes that can be targeted without harming healthy cells. Amongst the potential targets, researchers have validated their top candidate, COL11A1, through in vivo testing. As the momentum of scientific development into CAR-T therapy grows, building scalable manufacturing solutions is critical to enable widespread patient access and meet the growing demand for advanced therapies. Fantastic review on Circular RNA in Cancer by the Simon Conn group at the Flinders Health and Medical Research Institute. Circular RNAs (circRNAs) are gaining recognition for their significant roles in cancer biology, influencing oncogenesis at multiple stages. CircRNAs exhibit greater diversity (10x as the review states) and stability compared to traditional linear RNAs, making them promising candidates as biomarkers and for targeted therapies There is much ongoing research aimed at harnessing their unique properties for clinical applications.
