At 71, Marylou Barton was told her mesothelioma had spread, and hope for recovery seemed distant. But her daughters weren’t ready to give up. With the determination of her family, with support and funding from The Baker Street Foundation. Marylou joined a clinical trial at Memorial Sloan Kettering Cancer Center, where she received CAR-T. After her treatment, Marylou was given Keytruda® – an immunotherapy checkpoint inhibitor that supports CAR-T cells in sustaining their anti-cancer activity – at a hostpital in Pennslyvania. Now, nearly five years later and in remission, Marylou is enjoying time with family, traveling, and cherishing every moment of her retirement. Marylou’s story reveals the remarkable promise of CAR-T therapy, bringing hope when it’s needed most. Yet, as so many patients still encounter barriers to access, the real challenge remains in making these life-saving therapies accessible and affordable. 🔬 Our new data shows that IRO® produces CAR-T cells with a potent anti-tumor response – even at lower multiplicity of infections (MOI). CAR-T cells generated at MOI 0.25 and 0.5 in IRO achieved tumor-killing efficacy comparable to those produced with MOI 1.0 in the control platform. What does this mean? 🤔 The IRO produced CAR-T cells specifically targeted and killed CD19+ tumor cells while maintaining minimal non-specific activity. Importantly, IRO also met FDA guidance – keeping vector copy numbers (VCN) below three per CAR+ cell. 🔬CAR-T immunotherapy could be an option for patients with advanced prostate cancer. In an early clinical trial at City of Hope, researchers engineered CAR-T cells to target prostate stem cell antigens, a protein that is highly expressed in metastatic prostate tumors. Of the 14 patients treated, several showed reductions in prostate-specific antigen levels, with one patient seeing a drop of over 90% and tumor shrinkage lasting for eight months. The therapy was well-tolerated, with only mild side effects. As the momentum of scientific development into CAR-T therapy grows, building scalable manufacturing solutions is critical to enable widespread patient access and meet the growing demand for advanced therapies. 🔬 Early efficacy isn’t enough – commercial viability is key to widespread access to CGTs Recently on the Ori Spotlight Podcast, Jason C. Foster shared why – even in early clinical trials – developers need to assess if their therapy delivers enough doses, targets the right patients, and meets cost-of-goods targets. Without these factors, the path to commercial success becomes much harder and in some cases even impossible to achieve. Planning for scalability and cost-efficiency early on ensures therapies not only reach the market but are accessible to the patients who need them. 👑 Allan Peters, once a royal protection officer for King Charles, never expected his toughest battle would be with stage 4b lymphoma. After being told his cancer was terminal and his chemotherapy unsuccessful, Allan began preparing for the worst. However, everything changed when he was one of their first patients to be offered CAR-T therapy at University Hospital Southampton NHS FT. After a five-week stay (and a card from King Charles himself) Allan’s cancer was gone – two years later, he remains in long-term remission. Allan’s story shows the incredible potential of CAR-T therapy to offer hope when it’s needed most. However – with many patients still facing barriers to receiving the therapy they desperately need – the challenge lies in making these life-saving treatments approvable, accessible and affordable. At BioProcess International 2024, we shared exciting new data that highlights IRO®’s capacity to reduce manufacturing costs by up to 50%. When compared to a widely used, first-generation manufacturing platform, IRO delivered higher CAR-T yields at Multiplicity of Infections (MOI) 0.25 and 0.5 compared to the control at MOI 1.0. Thanks to its optimized transduction mixing protocols, IRO enhances both cell-cell and virus-cell interactions, boosting transduction efficiency and supporting higher cell densities. What does this mean? 🤔 By reducing the amount of lentiviral vector required, and improving transduction efficiency and cell expansion, IRO achieved the target CAR-T cell yield earlier – shortening overall process times and reducing COGs without compromising quality. 🔍 Want to learn more about the data?
