Exciting breakthrough in cancer therapy! Dr. Mitchell’s group at UPenn has developed a novel approach using chimeric antigen receptor (CAR) monocyte and macrophage therapies to target solid tumors more effectively than traditional CAR T cell methods. The key innovation? mRNA lipid nanoparticles (mRNA-LNPs) that enable precise, in situ engineering of CAR monocytes. This technique allows for temporary and adjustable expression of CAR, significantly minimizing the risk of affecting healthy tissues and simplifying the cell manufacturing process. However, finding lipid nanoparticles that specifically target monocytes and efficiently deliver mRNA inside cells has been challenging with conventional methods. Through innovative ionizable lipid design and high-throughput in vivo screening, scientists have discovered a new class of oxidized LNPs that naturally target monocytes and enhance mRNA delivery. An extensive comparison between oxidized and unoxidized LNPs revealed that oxidized ones show superior delivery to macrophages, without impacting T cells, due to their unique physical and chemical properties. Further, a specific oxidized LNP formula, C14-O2, was identified for its innate monocyte targeting capability. In a compelling proof-of-concept study, C14-O2 was used to engineer CD19-CAR monocytes directly within the body, achieving significant B cell elimination in healthy mice models. This pioneering research opens new doors for using oxidized LNPs in CAR macrophage/monocyte therapies, offering a promising new direction for treating solid tumors. #CancerTherapy #Innovation #CARmonocytes #Biotech #HealthcareInnovation
