💡 This week, we had the pleasure of hosting Karen McClymont and Gabriel Reines March from OracleBio as part of our spatial biology series. They presented two case studies: 1) The creation of a pathologist-validated and scalable image analysis workflow for a retrospective clinical study involving over 2,200 IHC-stained colorectal polyp whole slide images. 2) The use of Deep Learning apps to segment morphologically distinct cell subpopulations and the Phenoplex workflow to streamline the thresholding process for spatial phenotyping in multiplex IF-stained pre-clinical tissue. This recently published paper illustrates a protocol for synchronized wireless fiber photometry and video recordings in rodents during behavior! 📈 Fiber photometry is a powerful technique to monitor real-time neural activity by measuring fluorescence signals from specific neuronal populations. Traditionally, this was done using wired setups, limiting natural movement. Now, with wireless systems like TeleFipho (Amuza Inc.), neuronal circuits can be studied during complex, unrestricted behaviors. 📷 This study focuses on VIP+ interneurons in the dorsal CA1 hippocampus of mice, using the ANY-maze 𝗯𝗲𝗵𝗮𝘃𝗶𝗼𝗿𝗮𝗹 𝘃𝗶𝗱𝗲𝗼𝘁𝗿𝗮𝗰𝗸𝗶𝗻𝗴 𝘀𝘆𝘀𝘁𝗲𝗺 to record video and synchronize signals with photometry recordings. This setup ensures precise integration of behavioral data with neural activity, offering a flexible solution for researchers.
