Among these, the second stage, drug inhalation, is the most important and obscure. Using ambiguous verbal expression, the drug inhalation method is communicated to the patient using terms such as strongly, deeply, and slowly. Patients usually determine their optimal method of drug inhalation device independently, based on their own interpretation and understanding of the verbal instructions. This may make the precise inhalation using an inhalation device unpredictable. The Tokico Inhalation Monitor TM (TIM) was developed to resolve the unpredictability of this second (drug inhalation) stage. The TIM can simultaneously measure the inhalation flow rate, duration, and total volume, and display them on the screen in real-time. Rutin is a BCS class II drug having low aqueous solubility and therefore low oral bioavailability. In the present study, inclusion complex of rutin with hydroxypropyl-b-cyclodextrin were prepared by kneading method. Inclusion complex were characterized by 1H NMR, X-ray diffractometry (XRD), differential scanning calorimetry (DSC) studies, and Fourier transform infrared spectroscopy and evaluated for phase solubility study, saturation solubility and in vitro dissolution studies of plain drug (Rutin) and HPβCD inclusion complex. DSC and XRD study demonstrated that there was a significant decrease in crystallinity of pure drug present in inclusion complex (Rutin- HPβCD), which resulted in an increased dissolution rate of Rutin and 1H NMR studies strongly, confirmed that the inclusion complex has formed. The formulation so prepared were characterized for its disintegration, wetting properties, dissolution studies etc. Inclusion complexation results in improvement in solubility and dissolution rate. The inclusion complexation would be suitable method for dissolution and bioavailability enhancement of Rutin. The method is based on the oxidation of the studied drug by a known excess of potassium permanganate, followed by measuring the decrease in absorption (∆A) of KMnO4 in acidic medium at wavelength of 525nm. The detection limit is reported to be 1.01 and 1.06µg/mL showing a high degree of sensitivity. The proposed method was successfully validated according to ICH guidelines for the determination of esomeprazole and ciprofloxacin with a highly precise recovery and very low relative standard deviation.
