A great time at ELRIG UK Drug Discovery 2024 last week, hearing lots of new science, catching up with friends and colleagues and making new connections. A couple of the presentations that I found really interesting: 🧫 Therapeutic delivery of allogeneic mitochondria to promote cellular tissue repair and homeostasis by Dr. Alexander Schueller from cellvie. Fascinating biology; many challenges ahead, but could represent a completely new way to tackle tissue repair e.g. ischemic injury following transplant or cardiovascular events. 🧬 Achieving improved tumour penetration of antibodies by Zahra Jawad from CREASALLIS. Antibodies with protease cleavage-sites engineered into the hinge region - a simple but elegant approach that promotes the release of smaller antibody fragments into the tumour microenvironment.  📉 Reflections on the high attrition rate in Pharma R&D.  Cathy Tralau-Stewart from the Milner Therapeutics Institute.  Only ~10% of molecules that enter clinical trials gain regulatory approval; most fail for lack of efficacy in patients. Success rates can improve with a very careful focus on human biological evidence to select the best target, molecule and patient populations. Frustratingly, another frequent reason for project termination is simply a change in company strategy (e.g. disease area focus).  Surely there is something that the industry can do about that too? #ELRIGDD24 #ELRIG

There is an equally fascinating publication by @Ron Sender, Ron Milo and team on The total mass, number, and distribution of immune cells in the human body: https://lnkd.in/dT-RRHcb . These are fundamental data for developing immuno-oncology therapies where we try to direct the human immune system to fight cancer cells. Intuitively it is clear that in the fight of immune cells against cancer cells it matters what the numbers are. While this is well studied in vitro by exploring the E:T ratio (effector-to-tumor cell ratio) it is much more difficult to translate these findings to human: What is the concentration of immune cells in human tumors? How does it depend on tumor type? How does it vary between patients? How does it change over time? How high does it need to be to effectively destroy the tumor when being activated? I would love to see this work extended from healthy organs to disease in particular tumors.

What a great publication: The distribution of cellular turnover in the human body by Ron Sender and Ron Milo (https://lnkd.in/dMqpUcs6). Their research integrated ubiquity, mass and lifespan of all major cell types to achieve a comprehensive quantitative description of cellular turnover. Might sound unexciting to the unconscious reader but for Quantitative Systems Pharmacology #QSP modeller this is an invaluable resource to inform our mathematical models for new drug development.

🔬 We are thrilled to announce that our co-founder and scientific adviser, Michel De Waard, has contributed to a groundbreaking publication in the esteemed scientific journal BMC Complementary and Alternative Medicine and Therapies. 🎉 📚 Titled "Immunomodulatory effects of epiphytic Loranthus micranthus leaf extracts collected from two host plants: Psidium guajava and Parkia biglobosa" this study sheds light on the potential of natural immunomodulators in replacing current chemotherapy drugs with severe side effects. 🌟 Michel and collaborators’ meticulous research explores the immunomodulatory effects of Loranthus micranthus leaves extracts on innate and adoptive immune responses, as well as their impact on cyclophosphamide-induced myelosuppression in rats. 🔍 The findings reveal an abundance of pharmacologically relevant phytochemicals in both Loranthus micranthus leaf extracts, particularly in Psidium guajava. Moreover, the extracts exhibited promising immunostimulatory properties without adverse acute toxicity effects. 💡 Key discoveries include the extracts' ability to enhance total leukocyte count, differential leukocyte counting, and primary/secondary antibody titers, surpassing even the effects of the standard drug levamisole. The application of international reference pricing (IRP) systems International reference pricing (IRP), also known as external reference pricing, is a price control mechanism whereby a government considers the price of a medicine in other countries to inform or establish the price in its own country. IRP is used by, in particular, smaller countries with limited bargaining power or HTA capabilities to ensure that they do not pay more for medicines than other countries.  International price referencing may be used formally or informally to set reimbursement prices; at launch or on a regular basis; as the primary criterion for price setting or as one of the many inputs used to inform the pricing decision. IRP is extremely common – all EU and EFTA countries except Sweden and UK use external price referencing in one or the other form. Strategically integrating international reference pricing in your company But how does that work? Here are five steps to successfully integrate IRP management into a P&MA department’s daily business: 1. Establish an IRP library and keep it up to date 2. Develop a detailed pricing lexicon 3. Have in-depth knowledge of product characteristics 4. Create an IRP matrix 5. Incorporate a state-of-the-art, advanced analytics IRP model A good IRP Model contains all of the above; it takes all real-life IRP complexities into account to enable best-in-class management of the potential impacts of international reference pricing. This week we officially opened our #biobank of our trademarked UC San Diego HUMANOID: Human Organoid Center of Research Excellence (CoRE) PDOs to meet the growing demands of researchers (👇newsletter). Availability of these #phenotyped (and in some cases, also #genotyped), #PDOs that are #benchmarked in ways to show that they retain the #epigenetic drivers of disease phenotype and inter-patient heterogeneity, are expected to perform better as platforms for #drugdiscovery. We abide by, i.e., "less is more"; that way, the cell-autonomous features that are unique to diseases are likely to be reproducibly maintained. But #wait, there is #more (because we are #expanding #PDO #research to cover from the "#cradle to the #grave"), because it is never too early to impact #wellbeing if we start with the #mother and her #baby. 🚀 Exciting news! A new study from the Università degli Studi di Trieste explores the potential of microRNA (miRNA) profiles in neuron-derived extracellular vesicles (NDEVs) as predictive biomarkers for adverse drug reactions (ADRs) in children receiving ketamine. 🔬 This research, leveraging TAmiRNA GmbH's Next-Generation Sequencing (NGS)-based #miND® pipeline, identified specific miRNAs (miR-18a-3p, miR-484, and miR-548az-5p) as potential tools to predict ketamine-induced ADRs. 💡 This breakthrough not only advances our understanding of ketamine's side effects in children but also demonstrates the biomarker potential of organ-specific #EVs and their miRNA cargo.
