Bile acids (BA) are important metabolites and serve as signaling molecules, which are also involved in multiple cancer-related signaling pathways. Chenodeoxycholic acid (CDCA), a primary bile acid, has been reported to inhibit carcinoma cell proliferation in lung adenocarcinoma (LUAD). Lung cancer is the leading cause of cancer-associated death worldwide. Serum BAs were altered in lung cancer patients compared with healthy controls. CDCA inhibited LUAD cell proliferation, migration, and invasion. Furthermore, it promoted apoptosis (programmed cell death) in LUAD cells. It is postulated that altered bile acid metabolism can contribute to these diseases and bile acids have been identified as signaling molecules that regulate self-metabolism, lipid metabolism, energy balance, and glucose metabolism. Recent research suggests that bile acids could present a new opportunity for oncological researchers. This week, we sequenced RAB11B, short for Ras-related protein Rab-11B. RAB11B is a member of the RAB family of small GTPases, which are involved in the regulation of intracellular vesicle trafficking. RAB11B specifically plays a key role in the recycling of endosomes back to the plasma membrane, thereby maintaining cellular homeostasis and membrane receptor recycling. Therapeutically, targeting RAB11B has potential implications in cancer, as its dysregulation is often associated with tumorigenesis and metastasis. Additionally, RAB11B could be a candidate target for neurodegenerative therapeutics, where altered vesicle trafficking is a common feature in disorders like Alzheimer's disease. Our sequencing result demonstrates how Next-Generation Protein Sequencing™ data on the #Platinum® instrument can be used to sequence and characterize potential therapeutic targets, such as RAB11B. By analyzing the kinetic signatures from specific N-terminal amino acid recognizers, we identified 13 amino acids (F, Y, W, L, I, V, R, A, S, N, Q, E, and K), enabling identification of 4 peptides of RAB11B as shown in the figure. This study explores the impact of β-hydroxybutyrate (BHB) on clear cell renal cell carcinoma (ccRCC) cells under glucose deprivation, resembling a #ketogenic diet. Results indicate 𝗮 𝗱𝘂𝗮𝗹 𝗲𝗳𝗳𝗲𝗰𝘁, initially improving cell viability but eventually triggering apoptosis and suppressing unfolded protein response, highlighting potential therapeutic implications for managing ccRCC. Starving cancer cells by depleting specific amino acids, such as cysteine, can expose the vulnerabilities of cancer cells, leading to enhanced DNA damage and better responses to DNA repair-targeted and immune-based therapies. This strategy creates oxidative stress, disrupts glutathione synthesis, and amplifies reactive oxygen species levels, ultimately increasing cancer cell death. Judging by interactions, I thought you might enjoy another one. The main issue, i.e. that there is not enough evidence for nutritional therapy to make its way into guidelines, of course still exists - but this review makes an effort in giving a comprehensive representation about what we DO know, both on a macro- and micronutrient level. https://lnkd.in/e_5kmnRe.
