The findings of this secondary analysis of a randomized clinical trial suggest that the chemotherapy-free anti-ERBB2 strategy is feasible without being detrimental in terms of OS. The 50-gene prediction analysis of microarray signature could not help to identify the most appropriate patient population for this approach.

Overall survival rate estimated at 24 months was 52.4% with generally tolerated safety profile. Rakuten Medical recently completed a Type B End-of-Phase 2 (EOP2) meeting with the US FDA for the combination of ASP-1929 photoimmunotherapy and anti-PD-1 in the first-line recurrent HNSCC setting and discussed the key details for a multi-regional registrational study. This #ASH23 IsKia plenary article (https://clonoseq.co/rwm) states: While a similar percentage of patients in the two arms achieved a complete or stringent complete response, next-generation sequencing allowed us to see beneath the surface, exposing deeper responses in the Isa-KRd arm after consolidation, with 77% achieving MRD negativity at the 10⁻⁵ sensitivity threshold vs. 67% in the KRd arm (p=0.049). A “deeper dive” to the MRD threshold of 10⁻⁶ revealed that 67% of patients in the Isa-KRd arm achieved MRD negativity vs. 48% in the KRd arm (p<0.001). clonoSEQ was used to assess MRD in the IsKia trial, and can inform shared decision making in the clinical setting. Today Chemomab reported a new peer reviewed scientific publication that further validates the important role of our novel target CCL24 in the fibrotic and vascular manifestations of systemic sclerosis (SSc), a devastating and often lethal systemic rheumatic disorder that lacks disease-modifying therapies. In a 200+ cohort of SSc patients, those with high CCL24 levels showed higher mortality and greater disease severity across the skin, lung and vascular symptoms of SSc. Professor Francesco Del Galdo , lead author of the new publication and a systemic sclerosis expert at the University of Leeds, commented, “This is a crucial addition to the preclinical studies on the role of CCL24 in SSc, supporting the involvement of this profibrotic chemokine in key clinical manifestations of systemic sclerosis. The results provide further evidence that a CCL24-neutralizing antibody such as CM-101 could potentially be a valuable therapy for this devastating disease that has limited treatment options.” Chemomab's CM-101 program is Phase 2 ready and has an open US IND. We have successfully dosed the first of three patients in Cohort 8, the last cohort of the dose-ranging study in uMGMT #Glioblastoma Multiforme patients. The second patient has been enrolled and the treatment is planned.   •⁠ ⁠Thus far, the preliminary data indicate no dose-limiting toxicities related to Temferon have been detected in any of 22 treated patients. •⁠ ⁠Temferon-derived differentiated cells were evident within the peripheral blood 14 days after infusion and were still detectable at more than 24 months. •⁠ ⁠As of December 2023, preliminary data in uMGMT patients, the most aggressive form of GBM, show a 2-year Overall Survival (OS) of 25%; the historically reported data observed in uMGMT and methilated patients undergoing current standard of care is approximately 14% to 18%, respectively. Dr. Luigi Naldini, stated, “Several techniques, including RNA single-cell analysis, suggest that Temferon can reprogram the Tumor Microenvironment in patients similarly to what we had demonstrated in preclinical models and activate the immune cell infiltration towards mounting immune responses against the tumor.” Genenta Science (Nasdaq $GNTA)

A long waited trial from the group of Prof Bidard and Prof Pierga, but definitely worth the wait: the STIC CTC trial (full article at https://lnkd.in/dHrRWdz9) demonstrates that the use of CTC count as a tool for choosing between endocrine therapy (ET) and chemotherapy (CT) is non-inferior to the investigator's choice in terms of short and long-term clinical outcomes.
