Overcoming immunogenicity of a therapeutic cell product is one of the major hurdles to clear during Cell Therapeutic Development. One approach to overcoming this barrier is by using StemEdit Clinical Gene Editing Service to abrogate the expression of HLA in your therapeutic cells by knocking out the B2M and CIITA genes in the starting iPSCs. As part of our validation for the StemEdit service, we had created the B2M/CIITA double knockout in one of our iPSC lines. One crucial analytical technique that is highly relevant in the RNA field is capillary electrophoresis (CE). Compared with LC methods, CE-based methods show significantly better resolution of large RNA species and can be used to determine purity and integrity, marking a significant improvement in terms of resolution, reproducibility and automation capability; plus, state-of-the art instrumentation allows for temperature control to prevent the formation of secondary structures.
