In this lecture, Prof. Mark Levin will cover selected transformations discovered in his laboratory which address this challenge, enabling single-atom changes through the insertion, deletion, and/or exchange of single heavy atoms (C, N, O, etc.).

Delighted to start the new year with the release of our latest publication, where I had the opportunity to contribute to the computational analysis of optimized hits targeting the energy-coupling factor pantothenate transporter (ECF-PanT) in Streptococcus pneumoniae. Our work was centered on generating 3D compounds, which were optimized using dynamic combinatorial chemistry, and the detailed examination of two previously selected binding pockets of the ECF-PanT using molecular dynamic simulations (details in the Supplementary part). A key aspect of our work in the computational part also involved identifying the crucial interactions within the selected binding pocket. I am happy to announce our recent publication, a review article published in Advances in Protein Chemistry and Structural Biology by Elsevier. This paper explores the intricate catalytic versatility of metalloenzymes. Our research focuses on metalloenzymes, which utilize metal ions or cofactors within their active sites to facilitate a broad spectrum of biochemical reactions. The inherent properties of these metal ions—acting as Lewis acids or bases and redox centers—enable metalloenzymes to exhibit remarkable catalytic promiscuity. This study delves into the mechanistic underpinnings of this promiscuity, examining how metalloenzymes can catalyze various reactions beyond their primary functions. We highlight recent experimental and theoretical work to elucidate these phenomena. Have a look at the latest preprint on fragment screening with benchtop NMR. I am really glad that I could participate a (very little) bit to this story and I think there are many interesting ideas to look at in this article from Sami Jannin’s lab! This time it uses DNP and specific 13C labelling of the fragments to benefit from the relaxation and from the good spectral shift dispersion of 13C. This would enable to pool many compounds even at low field, alike 19F NMR. In this case, what is tracked is the T1 relaxation and not the T2 relaxation, and proves to be quite sensitive! Of course, 13C specific labelling remain relatively expensive to generate fragment libraries, but they are more and more articles describing high yield chemistry to insert stable isotopes from precursors. The chemistry described in the paper is very robust and high yield. Finally, I ran the math, and this would not add dramatically to the cost of the materials. We us much less of each fragment than of protein for the screening… Benchtop NMR fragment screening is becoming more and more widespread and this is excellent! Preprint: https://lnkd.in/evCjrrVi

Discover Our Impacts A paper titled “A Click Chemistry‐Based Artificial Metallo‐Nuclease” was published last year in Angewandte Chemie by Prof. Andrew Kellett, Dr Alex Gibney, Dublin City University and co-workers in SSPC, Chalmers University of Technology, and the Syddansk Universitet - University of Southern Denmark. 📌The paper was highlighted on the journal cover and was identified as a “hot paper” due to the significant findings reported. The paper reports the development of a new molecular scissors (or artificial metallo-nuclease) that can cut duplex DNA. The second article in the special collection SABIC 2024 is now online. Well this is the first one in Chemistry a European journal from Sankar Rath IIT Kanpur again. A great read.The article authored by Rath and co-workers, describes spin state changes facilitated by a series of sulfonate ligands ligated to diiron(III) porphyrins. It applies a multipronged approach: structural, spectroscopic, and computational, to better understand the factors controlling the spin states adopted by various porphyrin dimers.

It is part of a new field known as polariton chemistry.

Excited to share our latest publication in The Journal of Physical Chemistry C (https://lnkd.in/gNzyeecD). Our article on ‘methane pyrolysis in molten FeCl3/KCl catalyst’. This groundbreaking research reveals unique mechanism, where iron(III) takes up electron from several Cl ions and the charge-deficient Cl ions are active for catalysis. Here ab initio molecular dynamics (AIMD) simulations and the Nudged Elastic Band (NEB) method were used to study the catalytic activity of FeCl3/KCl melt for methane pyrolysis.
