The Adeno-Associated Virus (AAV) vector, AAV6.2FF, shows promising safety and scalability in expressing therapeutic monoclonal antibodies. In animal models, it demonstrated a safe profile and successful scale-up, indicating potential for generating immunity in specific populations. Drug discovery has seen many advances with the adoption of Ai/ML. The impact these tools can make are directly tied to good data measured by quality, quantity, and relevance of the data we feed our engine. At Pragma Bio have built our early success in making novel discoveries on the foundation of good data. Successfully completed the guided project on COVID 19 mRNA Vaccine Degradation Prediction provided by Coursera. This guided online project provided in-depth knowledge on the mechanisms of mRNA degradation, a crucial aspect in understanding the stability and efficacy of mRNA-based vaccines. Through this program, I gained valuable insights into the latest research and predictive models that can aid in the development of more effective vaccines and therapies. I'm happy to share our latest peer reviewed journal article discussing a surge in clinical trials involving genetically modified bacteria, bacteriophages (viruses that infect bacteria), and their risk assessments. Common disease indications under study include rare diseases, metabolic disorders, cancer, autoimmunity, and antibiotic resistant infections. This review article summarizes the United States Food and Drug Administration classification for investigational products (IPs) comprising bacteria or bacteriophages and provides an overview of clinical trials conducted to date involving genetically modified bacteria. The risk assessment for bacterial or bacteriophage-based IPs is discussed. The risk assessment process for bacterial or bacteriophage-based IPs is different from that of gene expression vectors and mammalian cells. Greater consideration must be given to the attenuating mutations affecting virulence, replication competency, antibiotic susceptibility, and persistence in the environment.
