Approved under the regulator’s accelerated pathway, Tecelra is also the first new synovial sarcoma therapy in more than a decade, according to Adaptimmune Therapeutics. Adaptimmune Therapeutics announced late Thursday that the FDA has approved afamitresgene autoleucel—now to carry the brand name Tecelra—for the treatment of unresectable or metastatic synovial sarcoma. Tecelra is the first engineered T cell therapy for solid tumor cancers in the U.S. and is the first new therapeutic option for synovial sarcoma in a more than 10 years, according to Adaptimmune. ICYMI: Next-gen heterobifunctional molecules are headed to the clinic. Halda Therapeutics (from the mind of Arvinas founder and PROTAC inventor Craig Crews) clinched a $126-million series B extension to start prostate and breast cancer trials of its Regulated Induced Proximity TArgeting Chimeras (RIPTACs). CSO Kat Kayser-Bricker told me that one half of a RIPTAC molecule engages with a “targeting protein” expressed in tumor cells, and the other half links up with an “essential protein” necessary for cell survival. Aiming for essential proteins has long been a goal of targeted oncology, but dose-limiting toxicities have hampered the approach. “But because we don’t interact in cells without that tumor-specific protein, we can have this very selective cell-killing mechanism," Kayser-Bricker explained. #TTUSInvents Targeted drug delivery has been among the fastest-growing fields in cancer therapy for decades. Advances in delivery technologies have led to the development of anticancer agents like antibody-drug conjugates (ADCs) and drug-loaded nanoparticles. Listeria monocytogenes (LM) is a Gram-positive intracellular bacterium that is being investigated in preclinical and clinical trials as a therapeutic cancer vaccine platform. The intracellular life cycle of LM, during which LM gains access to both endosomal and cytoplasmic spaces, significantly contributes to LM's versatility as an anticancer platform. However, LM is unexplored as a vehicle for therapeutic delivery directly into solid tumors and metastasis. Research out of the Department of Immunotherapeutics and Biotechnology at the Texas Tech University Health Sciences Center, by Dr. Laurence Wood, Dr. Devin Lowe and Wyatt Paulishak has resulted in the development of a novel mechanism for the tumor-specific delivery of anticancer agents using Listeria monocytogenes (LM) as a ‘living drug candidate’ and intracellular vehicle. This patent pending solution developed in collaboration with Dr. Michael VanNieuwenhze and James Lyu at Indiana University Bloomington leverages the invasiveness, immunogenicity, and inherent antitumor activity of LM to provide an efficient drug delivery vehicle with enhanced efficacy. Eisai Inks $1.5B Deal with Seed Tx for Molecular Glues Eisai and Seed Therapeutics announced a potential $1.5B+ collaboration to discover, develop, and commercialize novel molecular glue degraders for multiple undisclosed neurodegeneration and oncology targets. The deal combines Seed Tx’s focus on preclinical discovery of such targets and Eisai’s expertise in neurodegeneration and cancer. Exactly. Look at therapeutic regimens in Oncology - or any disease for that matter. How many are strictly mono therapies? In my 25 years or drug discovery and development - the aspirational goal has always been monotherapy. How often have we actually achieved this. Most effective treatments are a multipronged/poly-mechanistic approach. Systems biology is complex and filled with multiple redundancies. Biology and therefore Diseases are not simple binary in off switches. 😁😁
