Orexin receptors are proteins located on the surface of nerve cells in the brain and other tissues of the body that respond to orexin, also known as hypocretin. Orexin is a neuropeptide that plays a crucial role in regulating sleep, wakefulness, and feeding. There are two main types of orexin receptors, known as OX1 and OX2, which are members of the G protein-coupled receptor (GPCR) family. In this study, the homology structure of OXR1 was obtained using Modeller 10.4, based on the PDB model 6TO7. The CHARMM-GUI server was used to embed the receptor in a POPC bilayer membrane. The orexin 1 peptide was obtained from the PDB model 1CQ0. I love this kind of medical discovery: the microcosmos. It is obvious now that there are many organelles and cell functions of the human body that we don't know exist yet. We often wonder about the macro cosmos and don't pay much attention to the awe that exists in the microcosmos. Many things are to be discovered yet. I am excited about this news.
🌎 The World of Migrasomes 🌎 The migrasome is a newly discovered organelle produced by migrating cells. As cells migrate, long and thin retraction fibers are left in their wake. On these fibers, we discovered the production of a pomegranate-like structure, which we named migrasomes. The production of migrasomes is highly correlated with the migration of cells. Currently, it has been demonstrated the migrasomes exhibit three modes of action: release of signaling molecules through rupturing or leaking, carriers of damaged mitochondria, and lateral transfer of mRNA or proteins. Some years ago, I was intently studying time-lapse movies of fluorescently labelled co-cultured cells that transferred contents between each other. I had seen that large amounts of material were transferred, especially from fibroblasts to cancer cells, but could not easily see how it was happennning. The recordings were ‘con-focal stacks’, so I could focus up and down the cells to see at what level events occurred. Doing so, I noticed cell debires was clinging to the culture surface in a net-like pattern, and for a day or two I thought that perhaps the transfers I was seeking might be just the slow accumulation of these fragments as cells migrated about. But look how I might, I couldn’t see any clear uptake of these net-distributed particles. It was then that I first stumbled across a dramatic and unambiguous episode of what I later came to call ‘Cell-Projection Pumping’ (CPP), which we subsequently demonstrated accounted for the transfers I had been studying and also for the changes in cancer cell behaviour we had seen before. I put the ‘net of debris’ aside, and focussed on what I was looking for, but now see how on that occasion, I had failed to follow an interesting scientific observation - what is now called ‘the migrasome’. It is in some manner ironic, because I am quite certain that many other workers have noticed incidental episodes of CPP, but have like I did for the migrasome, not really ‘seen it’ because their interest lay elsewhere. I wonder how many other interesting discoveries are briefly noticed and then ignored, waiting for someone else to pick them up and chase?
