I feel on cloud nine to announce the publication of our article entitled “Selective Inhibition of indoleamine and tryptophan 2,3-dioxygenases: Comparative Study on Kynurenine Pathway in Cell Lines via LC-MS/MS-based Targeted Metabolomics” available under open access licence for everyone who may be interested in. 😄 Here you may find the direct link: https://lnkd.in/gyutZR78 It concerns a validated LC-MS/MS method to quantify the “kynurenines”, which are the major tryptophan catabolites, in cellular supernatants. Our method was successfully applied to assess the in vitro potency of IDO1 selective inhibitors in early drug discovery phases and to obtain vital information about our cell models (A375, U87, MDA-MB-231 cell lines). All the obtained info will be exploited to support the biological characterization of novel inhibitors targetting IDO or TDO. A deep dive into #Pinetree's platform suggests that: 1) The #AbReptor platform utilizes Neurophilin-1 Receptor (NRP1) as the effector protein to promote internalization and degradation, a mechanism similar to #LYTAC; 2) It originates from Dr. Yong-Sung Kim's lab in Ajou University on the study of #NRP1-Tumor penetrating peptides; 3) NRP-1 can interact with integrin β3 to overcome EGFR resistance in multiple #KRAS-mutants NSCLC, potentially via NRP-1-triggered complex co-internalization and degradation; 4) Pinetree further develops a scFv-based NRP1 binder optimized for internalization and degradation purposes.
