Forbion portfolio company AZAFAROS B.V. has announced that data from the ongoing double-blind, placebo-controlled Phase 2 RAINBOW study investigating its lead asset, nizubaglustat in patients with Niemann-Pick disease type C (NPC) or GM2 gangliosidosis, were presented at the Society for the Study of Inborn Errors of Metabolism (SSIEM 2024 Annual Symposium) in Porto, Portugal. The results of part one of the study, designed to determine the safety, #pharmacodynamics, and #pharmacokinetics of Azafaros' lead asset nizubaglustat, demonstrated the compound had a positive safety profile and was well-tolerated in the 13 participants in the study. All patients are now enrolled in the extension phase of the trial, where initial unblinded data on 10 patients showed 8 had improved or stabilized SARA (Scale for the Assessment and Rating of Ataxia) scores. Additionally, a reduction in seizure frequency was observed for the participants who had experienced daily seizures before recruitment into the study. Data from the RAINBOW study will be used to inform the target dose for Azafaros’ planned Phase 3 pivotal studies, due to be initiated 2025. Radiolabeled “Absorption, Distribution, Metabolism, and Excretion” (ADME) studies help understand how organisms handle a candidate drug. While in most cases, nonclinical ADME data from animal models predict clinical excretion routes, in some cases, drug metabolism and excretion routes are substantially different - highlighting the relevance of conducting human ADME studies. Traditionally Arnica montana L. is indicated for treatment of blunt injuries like strains, bruises and contusions that are physiologically accompanied by local inflammation and edema, including activation of the NF-kB pathway and the release of pro-inflammatory prostaglandins and leukotrienes that are produced by cyclooxygenase-2 (COX-2) and 5-lipoxygenase (5-LO). Arnica flowers (A. flos) contain compounds, e.g. sesquiterpene lactones like helenalin or dihydrohelenalin that are known anti-inflammatory compounds. Join Johann Röhrl, PhD, Senior Manager Preclinical Development, WELEDA UK at this 30-mintue workshop taking place at the IPM Congress on Saturday 8 June, to evaluate the anti-inflammatory efficacy of a hydroethanolic extract from A. flos. Register to attend for just £25 per day. Excited about the final day of the #APAC track of #RinPharma today! 🚀 Please dial in via rinpharma.com 💻 Yesterday we had a cancellation of a talk for today's schedule, so I came up with a replacement talk quickly: "R(omeo) and Julia - A Love Story by #openstatsware" I presented this once before at the PSI webinar in April this year (https://lnkd.in/g_8nmgtz), but I think it is worth repeating. Looking forward! #rstats #julia #biostatistics #jointmodels #safetysignaldetection

New work from the lab: Cryo-EM structures of the cytotoxic food-borne pathogen Clostridium perfringens enterotoxin bound to its human receptor, claudin-4. https://lnkd.in/evU5uHpZ This is a structure we’ve been chasing since 2019 by crystallography that had stalled, but was resurrected by cryo-EM.

Join Anna Krause during Venom Week 2024, which starts on June 5th in Durham. Anna will present the results of Phase I clinical study of Marimastat, a potent inhibitor of snake venom metalloproteinases (SVMPs) Marimastat showed excellent efficacy in in vivo models of snakebite envenoming, protecting mice from lethal doses of various venoms of viperid snakes. It has the potential to become a safe, efficient, and affordable oral drug for snakebites, protecting victims against tissue damage and hemorrhage and, ultimately, from limb amputation or even death. To the best of our knowledge, Pikralida is the only company in the world with a current regulatory dossier, GMP-quality Marimastat, and a drug product suitable for human use.
