The SampleStream platform is ideal of interfacing affinity capture workflows to mass spectrometry and this work by Seckler et al. demonstrates a few of the reasons why: 1.) Concentration - The SampleStream platform is a molecular trap, so the entire IP elution volume can be injected and concentrated into a fixed elution volume. The result of this concentration is improved sensitivity for challenging targets. 2.) High Throughput - Affinity purified samples are relatively simple and in most cases current HRAM MS system are capable of resolving all of the components without the need for LC. The SampleStream platform gets samples to the MS fast (1-3 min per sample depending on target MW) so you can get data back to your stakeholders fast. 3.) Buffer Exchange - Affinity selection workflows sometimes require the use of elution buffers that are not mass spec compatible. The SampleStream platform rapidly exchanges many of these components so no further processing is required.

The samples were then processed via SampleStream-Mass Spectrometry and compared to LC-MS analysis of the same samples. While this early version of SampleStream suffered a slight sensitivity deficit to the LC-MS approach, it provided 7x the throughput and reduced CVs by over 50%. The simplicity and speed of the platform eliminates many of the failure modes associated with liquid chromatography and the improved speed reduces the impact of instrument drift.
