A new study has developed a model system to investigate the function of CK1 variants in the Wnt/β-catenin pathway. A molecular dynamics study from the collaboration between AO Research Institute Davos (ARI) and Dalle Molle Institute for Artificial Intelligence ( USI Università della Svizzera italiana-Scuola universitaria professionale della Svizzera italiana (SUPSI) ) identified a possible physical interaction between the #Glucocorticoid #Receptor (GR) and #SOX9. This interaction appears modulated by #dexamethasone (DEX), making it more stable. ❓ We currently don't know what the function of this complex is, but SOX9 carries the GR coactivator motif LXXLL which is partially involved in the binding process. Also, we hypothesize that SOX9 can be "sequestered" by GR in the presence of DEX, preventing its dimerization and transcriptional activation of its targets - including SOX9 itself. This mechanism might explain at least partially why DEX induces SOX9 downregulation during in vitro #osteogenic differentiation of human bone marrow MSCs. 🧐 The implications of our findings can be manifold, and the upcoming steps are focused on unravelling the function of this complex.
