The Methoxy Magic: Exciting progress in #HDAC11-selective inhibitors! This study from #Harvard Medical School introduces #PB94, a potent and selective HDAC11 inhibitor with promising drug-like properties. Unlike previous #inhibitors, PB94 is designed for effective blood-brain barrier penetration, a critical factor in neurological disorders. "The discovery of #AZD4747", "Potency is sacrificed for CNS exposure."  Researchers from #AstraZeneca revealed their KRASG12C inhibitor with improved CNS exposure. The article discusses the discovery of AZD4747, which is a potent and selective inhibitor of the #KRASG12C protein. #Researchers used the strategy of removing the essential #pyrimidine ring from the structure of the compound to make it smaller and more lipophilic, following the #Lipinki rule of 5 for CNS #drugs. This modification led to a CNS penetrating compound with decent potency. In animal studies, it showed promising CNS availability. Modifications at positions C3 and C17 of the #steroids have consistently shown intriguing results. In this study, a novel approach is revealed to treat castration-resistant prostate #cancer. The approach involves degrading the #androgen receptor protein (AR) and inhibiting the #CYP17A1 enzyme responsible for androgen synthesis with a compound called #YXG158. In this study, the authors conducted further optimization of compound (6), originally derived from galeterone (5), resulting in the development of YXG-158 (23-h). YXG-158 (23-h), emerged as a promising #bifunctional AR #degrader and CYP17A1 inhibitor to treat #enzalutamide-resistant CRPC. YXG-158 displayed potent AR degradation and moderate CYP17A1 inhibition in vitro, along with significant antiproliferative activity in cancer cells. Importantly, 23-h did not inhibit #hERG and showed favorable #pharmacokinetic profiles in rats. The direct conversion of inactivated #sp3 #alcohols into #oxetanes. Oxetanes are special types of molecules with unique properties that have led to important progress in medicinal chemistry. However, making these molecules in the lab is still quite difficult, which limits their use in real-life applications. In this research paper, the scientists came up with a new way to make oxetanes from regular alcohol molecules. This new method is versatile and can be used in late-stage chemical modifications. They used this approach to create a synthetic steroid derivative, which previously required at least four different steps to make. Now, with this new method, they can make it in just one step.
