It's Fluorescence Friday again! Today we share a beautiful montage of two-color dSTORM images of nuclear pores, a large protein complex found that mediates the selective transport of molecules across the nuclear envelope in eukaryotic cells. 🔬 Using ONI's Discovery Kit: dSTORM in cells, Nup96 (green) and Nup98 (magenta) were labeled to show the outer and inner rings of the nuclear pores. U2OS cells expressing mEGFP-Nup96 fusion were stained with primary antibodies against GFP and Nup98 nuclear complex protein, followed by secondary antibodies conjugated to AZ647 and CF583R, respectively. Simultaneous two-color imaging of both channels was performed using the Nanoimager. Nuclear pores are as small as 100 nm in size! 🔬 The Discovery Kit together with ONI's analysis tools on CODI can extract information, such as the radius of each nuclear pore protein. In this case, the Nup96 radius peaks at 60 nm, and the Nup98 ring radius at 40 nm. The new HCN SB DNP probes are available for LT-MAS below 100 Kelvin, with high-resolution MAS even at low temperatures. Bruker is also developing high-field gyrotrons for microwave frequencies of 658 GHz to 790 GHz for solid-state DNP-NMR at 1.0 and 1.2 GHz. Nuclear transport is a key #biologicalprocess involved in physiological and pathological events, which can be regulated by post translational medications of cargoes. In 2022 NKS funded a project called POSEIDON (Position-sensitive detectors for nuclear fuel imaging). In the project new types of detectors were used to measure spent nuclear fuel elements. A new peer-reviewed article has been published where mock-up fuel bundle was used to test the detector. New nuclear-electronic orbital (NEO) features: NEO-SCS-RIMP2 and NEO-SOS'-OOMP2! These methods provide improved accuracy over NEO-MP2 by compensating for the lack of orbital optimization, yielding results that are competitive with NEO-CCSD methods while retaining MP2's favorable scaling. NEO methods account for nuclear motion in quantum chemistry simulations, allowing key nuclear quantum effects to be incorporated! New publication in eLife of the Jordan Lab in collaboration with the IBMB Imaging Platform showing that some human histone H1 variants are universally enriched at the nuclear periphery close to lamin while others are distributed throughout the nuclei, with the particularity of H1X being enriched within the nucleoli, with putative distinct participation on chromatin and nuclear structure organisation. https://lnkd.in/dceQ-xaU

On Cosmological Low Entropy After the Big Bang: Universal Expansion and Nucleosynthesis and Gary the Brave Neighbour Charlie F. Sharpe , Luke A. Barnes and Geraint F. Lewis Abstract We investigate the sensitivity of a universe’s nuclear entropy after Big Bang nucleosynthesis (BBN) to variations in both the baryon-to-photon ratio and the temporal evolution of cosmological expansion. Specifically, we construct counterfactual cosmologies to quantify the degree by which these two parameters must vary from those in our Universe before we observe a substantial change in the degree of fusion, and thus nuclear entropy, during BBN. We find that, while the post-BBN nuclear entropy is indeed linked to baryogenesis and the Universe’s expansion history, the requirement of leftover light elements does not place strong constraints on the properties of these two cosmological processes.
