Metabolic network analysis of pre-ASD newborns and 5-year-old children with autism spectrum disorder Metabolism is strongly linked to child development. Children with ASD exhibit distinct metabolic profiles that vary by age, sex, and symptom severity. These profiles, in part, appear to drive the neurodevelopmental aspects of ASD. This study employed classical and network metabolomic analysis to identify key metabolic differences in 5-year-olds with ASD. These differences involve phospholipid metabolism, fatty acid oxidation/acyl-carnitines, and sphingolipid metabolism. Notably, 80% of the metabolic impact stems from shared pathways identified in both pre-ASD newborns and affected children. The ASD metabolic phenotype is characterized by decreased anti-inflammatory/antioxidant molecules (glutathione, carnosine, folate, CoQ10) and a coordinated increase in stress metabolites (lactate, glycerol, alanine, threonine, cholesterol, ceramides). Importantly, the magnitude of these changes worsens with age.
