Existing methods for cell classification rely on heuristics or user parameters to determine the number of clusters, impacting cluster resolution and replicability across datasets. 💡 𝐃𝐮𝐧𝐞 is introduced as a method to optimize the balance between cluster resolution and replicability by merging clusters within partitions. It has shown superior performance in replicability and concordance with ground truth compared to existing techniques on various datasets. 🔑 𝐃𝐮𝐧𝐞's cluster refinement method enhances clustering analysis robustness and decreases parameter tuning dependency. I'll be giving a talk on "Protein folding and what it means for drug discovery" at PyData London 2024 on Sunday, June 16th. I'll be giving the talk from the perspective of someone who's worked as a pharmacologist and as a software engineer rather than from the perspective of a bioinformatician or a cheminformatician. I'm planning to work backwards from a well-known receptor/ligand relationship and then see how this could have been discovered by using virtual screening, protein folding and the like and hopefully give attendees a sense of how to practically go about this. I’m excited about Illumina’s recent acquisition of Fluent Biosciences and their innovative, largely instrument-free, emulsion-based single-cell sequencing method, PIP-seq. I have great confidence in Illumina’s R&D team to elevate PIP-seq, potentially making it a strong competitor against established methods from 10x Genomics, Parse, and Scale. However, there are two key areas that I think need further attention: 1. 𝗘𝗻𝗵𝗮𝗻𝗰𝗲𝗱 𝗦𝗮𝗺𝗽𝗹𝗲 𝗙𝗶𝘅𝗮𝘁𝗶𝗼𝗻 𝗦𝘂𝗽𝗽𝗼𝗿𝘁: The current documentation for the PIP-seq DSP/Methanol fixation protocol is quite basic and needs improvement. Storing and sequencing millions of cells from multiple different samples is still an underestimated logistical challenge. 2. 𝗛𝗶𝗴𝗵-𝗧𝗵𝗿𝗼𝘂𝗴𝗵𝗽𝘂𝘁 𝗦𝗮𝗺𝗽𝗹𝗲 𝗣𝗿𝗼𝗰𝗲𝘀𝘀𝗶𝗻𝗴: As other single-cell methods have introduced automated workflows to minimize hands-on time, it’s crucial to focus on efficiency.
