#METABOLITE #GPCR sensing - how #ketones/beta-hydroxybutyrate; mono-methylfumarate (the #MS drug), and #niacin work. Amazing how the puzzle of 30 (thirty) new #HCAR2 structures when pieced together by Asli (Aslihan Shenol Iliyaz), Ricardo Tenente, Michael Lückmann, and Thomas Frimurer shows the whole beautiful picture of the electrostatically guided, complex agonist binding path into a totally occluded orthosteric site in the middle of the receptor; how an #agoPAM surprisingly works by controlling the dynamics of the entry/exit gate; and how a two-way allosteric mechanism connects G protein with agonist binding. G protein binding ALONE stabilizes ALL major conformational changes - including extracellular loop 2b - except only the induced-fit anchoring of agonist in the orthosteric pocket.
