𝗦𝗮𝗺𝗽𝗹𝗲 𝘀𝗽𝗶𝗸𝗶𝗻𝗴 - 𝘄𝗵𝗮𝘁'𝘀 𝗶𝘁 𝗮𝗹𝗹 𝗮𝗯𝗼𝘂𝘁? Spiking is a common technique for method validation in analytical chemistry. It helps to identify matrix effects and determine the recovery of an analyte or the reliability of the analytical method. It means to add a known quantity of a substance, which should be close to or identical to the concentration of the analyte of interest, in the sample matrix. Typically, the added spike is different from the analyte of interest, Consequently, the original sample as well as the spiked sample are then analyzed. The latter will show a larger analytical response than the original sample due to the additional amount of analyte. The relation of the spiked to the original result, considering the spike added concentration, gives the % recovery. Typically, acceptable spike recoveries have a wider specified range than recoveries of calibration verification standards.
