In collaboration with Waters Corporation, Similis Bio demonstrated an automated Protein A purification and LC-MS analysis of bioreactor samples. The goal? To screen biosimilar monoclonal antibody (mAb) candidates and compare them to the innovator product during process development. The method proved sensitive and precise, allowing quantitative assessment of N-linked oligosaccharides and other post-translational modifications (PTMs). By evaluating these quality attributes early in clone selection, biopharmaceutical organizations can reduce subsequent development costs. This application note demonstrates a two-dimensional liquid chromatography/quadrupole time-of-flight mass spectrometry (2D-LC/Q-TOF) approach to identify the free drug content of an ADC sample. The method combines size exclusion chromatography (SEC) in the first dimension (1 D) and reversed-phase (RP) separation in the second dimension (2 D).
