Redefining #PFAS is an unfortunate, legal trend that seems to be spreading across the USA, in opposition to the opinion of OECD Environment, many regions of the world, and protests by hundreds of scientists. The initiative seems specifically designed to prevent F-gases and other precursors of trifluoracetic acid (#TFA) from being regulated as #PFAS, despite the rapid and irreversible increase of #TFA in our blood, groundwater and even trees. The rapid accumulation of #TFA shows little sign of slowing, and constitutes a planetary boundary threat (see https://lnkd.in/d9p8PgDw). On the table is Indiana House Bill HB1399 (https://lnkd.in/dbMHHdZE ). To counteract this trend, a petition has been organized by the Green Science Policy Institute, where scientists with expertise in #PFAS can sign their support for the #OECD definition of #PFAS of a substance having "“at least one fully fluorinated carbon”. A novel study by Wei et al. introduces an advanced deconvolution method assisting in annotating PFAS homologues in multisample Studies. Using a SCIEX X500R QTOF system, the authors compare the performance of Data Dependent (DDA) vs Data Independent acquisition (SWATH) as a tool for the discovery of novel PFASs. The stochastic nature of DDA along with the intensity bias has long been the driver for the adopting of SWATH acquisition in multiple fields of science (as the authors recognize). Nevertheless, the chimeric nature of SWATH spectra does pose a challenge for the direct identification of PFAs. In this publication, the authors develop “A Novel Nontarget Strategy Based on the SWATH-MS Deconvolution Method” consisting of 2 parts: spectral deconvolution followed by the peak picking and ion annotation for PFAS homologues. Given that the file format and data export form after deconvolution was deemed inappropriate for streamlined compound identification, the authors developed their own tool to automate the process. They evaluated the application of SWATH-F to gradient spiked samples and real population serum samples, having obtained better results for SWATH-F with 276% improvement in identification.
