📢Continuous PAT in Bioprocessing 3: Sterile Sampling Sterile sampling kits and sterile sampling probes are widely used for safe and ⚡contamination-free medium extraction⚡ in bioprocesses. This provides real-time information, assists the improvement of the overall efficiency of the bioprocess and generates data for AI-based models. Sterile sampling can be facilitated by either pulling out sample through sterile filter membranes, check valves, septum-based solutions or sterile connector systems and further. Repeatable-use sterile sampling probes offer the highest degree of automation and can thus interface sensor components that automatically provide valuable data with short intervals between sample draw-outs and even continuous flow read-out. Contamination-free repeatable-use sampling: ⁉Sterile sampling probes ensure the sterility of closed cultivation systems by employing either micro filter membranes or probe-tip mounted check valves. After extraction through these filters or valves, reactor medium sample is pulled out and analyzed outside immediately of the bioreactor. Further options for continuous, repeatable-use sampling present themselves by separating the fluidic line of an extraction port with a peristaltic pump roller, by a check-valve or a combination of both. Additional measures to ensure the safety of the reactor can encompass sterile air flushing, introduction of cleaning agents, localized heating or UV-irradiation for automated sanitation. Our enzymatic-based biosensor components can be directly interfaced with the outlet of such probes, giving immediate readings of Glucose, Lactate, Glutamine (and others) right at the point of exit without any time delay or manual handling required 🎯. 👉Real-Time Adjustments to target optimal culture conditions: Repeated, automated monitoring allows for real-time adjustments to be made, ensuring that the bioprocess remains within desired parameters range. Having dedicated biosensor components interfacing a reactor provides the opportunity for frequent automated sampling and analysis. With direct measurement at the reactor site a hands-off approach is facilitated, allowing for scalable application with minimal geometrical footprint, and the capability to automate reactor oversight and control minute by minute 24h per day, 7 d a week. 👉Components for automated at-line monitoring Jobst Technologies provides enzyme-based biosensors suitable for at-line monitoring of Glucose, Lactate, Glutamine, Glutamate, and others✅. One effective solution to generate online readings of metabolic conditions is the use of microdialysis probes that can withstand autoclaving. These probes can be pre-mounted in single-use bags, T-flasks or smaller scale disposable reactors via LUER Lock ports and other mounting configurations. ⁉ During operation, a continuous stream of transport fluid in the single µl/min range flows through the probe. It passes a semi-permeable membrane that allows small molecules (like metabolites) from the surrounding reactor medium to diffuse into the transport fluid but poses a tight barrier for microbial contaminations.
