🔎 A very interesting study was published on the Journal “Environmental Health Perspectives” in august, titled “ Effects of Early Life Exposures to the Aryl Hydrocarbon Receptor Ligand TCDF on Gut Microbiota and Host Metabolic Homeostasis in C57BL/6J Mice”. ✔️ 2,3,7,8-tetrachlorodibenzofuran (#TCDF) is a widespread persistent organic pollutant (#POP) that is a byproduct of waste incineration, metal production, and fossil-fuel and wood combustion. TCDF accumulates in the food chain, and humans are primarily exposed through consumption of high-fat foods, such as meat, dairy products and some fish, while neonates are exposed through breast milk consumption. 🐁 The effects of this chemical were assessed on two groups of four-week-old mice – test and control group. Test group was fed 0.46 µg of TCDF for 5 days. Data on the health of mice was collected immdiately after the 5 days treatment period and 3 months after the last dose. In humans, these time points are equivalent to an infant and a young adult. TCDF-exposed mice exhibited lower abundances of A. muciniphila, lower levels of cecal short-chain fatty acids (SCFAs) and indole-3-lactic acid (ILA), as well as lower levels of the gut hormones glucagon-like peptide 1 (GLP-1) and peptide YY (PYY), findings suggestive of disruption in the gut microbiome community structure and function. ❗ Moreover, researchers transplanted microbiome from the TCDF-treated animals to germ-free (GF) mice, and measured their health outcomes. Importantly, microbial and metabolic phenotypes associated with early life POP exposure were transferable to GF recipients in the absence of POP carry-over. This research suggests how the exposure to 'forever chemicals' during early childhood may be contributing to the recent epidemic of metabolic disorders, including obesity and type 2 diabetes among adults, due to their ability to permanently disrupts the gut microbiome in mice. 🔥 #ECHA has published a new Assessment for Regulatory Needs focusing on eight structurally similar substances characterized by esters from the dicarboxylic acid moiety. The group includes seven substances with full registrations and one with C&L notification. ➡️ These substances are primarily used in industrial applications such as plasticizers in polymer production, laboratory use, lubricants, metalworking fluids, and as intermediates. Based on the current data from in vivo and QSAR studies, hazards like carcinogenicity, mutagenicity, reproductive toxicity, endocrine disruption, and others are unlikely for all substances in this group. There is currently no data indicating endocrine-disrupting activity for any of the substances. ▶️ #ECHA recently published an Assessment for Regulatory Needs, focusing on 13 structurally similar substances characterized by the lactic acid moiety. This group includes seven substances with full registrations and others with C&L notifications or pending registration. These substances are widely used in everyday products like cleaning agents, #cosmetics, #biocides, #lubricants, and #fragrances, leading to significant exposure and environmental release potential. 🔎 Key findings: 🌀 Hazards such as mutagenicity, carcinogenicity, reproductive toxicity, and endocrine disruption are considered unlikely for all substances in the group. 🌀 Skin sensitization is not expected, apart from one substance (List 606-097-1), which is self-classified for skin sensitization. 🚨 The European Food Safety Authority (EFSA) has released a new evaluation on the #thyroid-related effects of various #pesticides active substances and metabolites, grouping them into Cumulative Assessment Groups (#CAGs). These CAGs focus on indicators of hypothyroidism and C-cell effects like hypertrophy, hyperplasia, and neoplasia. 🔎 Key findings: 🌀 18 substances and three metabolites were linked to hypothyroidism (CAG-TCF), while two substances were associated with C-cell abnormalities (CAG-TCP). 🌀No substances showed effects in both CAG-TCF and CAG-TCP categories. 🌀LOAELs for CAG-TCF ranged from 1.1 to 4570 mg/kg bw/day, and NOAELs from 0.11 to 457 mg/kg bw/day. For CAG-TCP, LOAELs ranged between 5 and 7.31 mg/kg bw/day, and NOAELs between 0.5 and 1.46 mg/kg bw/day. This report offers important insights for risk assessments of pesticides and #Endocrine #Disruptors (#ED), and confirms consistency with previous evaluations for ten substances. #ToxHub #EFSA #Consultancy #Publication #PPP

🚨 European Food Safety Authority (EFSA) has just released a comprehensive review on using New Approach Methodologies (#NAMs) in assessing #nanomaterials (#NMs) in #food and #feed, building on their 2021 guidance, which emphasizes Integrated Approaches to Testing and Assessment (#IATAs) to fill data gaps and assess small particle fractions. ▶️ In particular, 30 frameworks for NMs were reviewed, covering Adverse Outcome Pathways (#AOPs), IATAs, and physicochemical characterizations. EFSA's three recommendations to advance regulatory use of NAMs: 1. Engage with regulatory experts to discuss mature NAMs; 2. Test NAMs in nano-specific case studies to address challenges; 3. Focus on developing and standardizing NAMs across regulatory bodies in the EU. 💫 This report is a crucial step in advancing NAMs for more robust and informed risk assessments in the food and feed sectors. 💥 The European Food Safety Authority (EFSA) has released an updated Scientific Opinion on Tetrabromobisphenol A (#TBBPA) and its five derivatives in food, revisiting findings from the original assessment issued in 2011. ➡️ TBBPA belongs to the class of Brominated flame retardants (#BFRs), anthropogenic chemicals, which are used in a wide variety of consumer/commercial products to improve their resistance to fire. Compounds from the BFRs groups have been found in the environment, food and in humans, leading to #bans on their production and use in certain products. Neurotoxicity and carcinogenicity were considered as the critical effects of TBBPA in rodent studies. 🔥 The available evidence indicates that the carcinogenicity of TBBPA occurs via non-genotoxic mechanisms. Taking into account the new data, the CONTAM Panel considered it appropriate to set a tolerable daily intake (#TDI) of 0.7 µg/kg bw/day. 🆕 From April 2026, Undecafluorohexanoic acid (#PFHxA), its salts, and PFHxA-related substances will face restrictions across the European Union due to their hazardous properties, particularly their classification as #PMT (Persistent, Mobile, and Toxic). ➡️ While PFHxA itself isn't registered or used in the #EU, its related substances and the ammonium salt are. These compounds, used in tonnages ranging from 1 to over 100 tonnes annually, play a major role in industries like: 📦 Paper & cardboard (food contact materials); 👕 Textiles ; 🔥 Firefighting foams. This regulatory change marks a significant step toward reducing the environmental and health risks posed by PFAS. #ChemicalSafety #Sustainability #EU #EnvironmentalProtection #IndustryUpdates Follow us for regulatory updates and scientific insights! 💻www.toxhub-consulting.com 📩 info@toxhub-consulting.com
