🌼 Explant Selection, Sterilization and Callus Induction 🌼 Hi everyone! I’m excited to share with you about my productive day yesterday at Agro-Biotechnology Institute (ABI), National Institutes of Biotechnology Malaysia. 3 main activities have been done yesterday which are #explantselection, #sterilization and #callusinduction. 1. Explant selection - The species that I’m experimenting with is Trichantera gigantea or commonly known as Ketum Ayam. We chose its leaves as explant for this experiment. In tissue culture, it is crucial to choose the best and healthiest explant. Due to the morphology of the leaves having short hairy structure, it is a bit challenging to find one that is completely healthy and have no infection. However, we managed to find and select some for our next step. 2. Explant Sterilization - Then, the leaves are brought into the lab for sterilization. It is very important to take these 2 things into consideration, first, the concentration of the sterilizing agent and second, the period of time where the explant are in contact with the sterilizing agent. Usually, the higher the comcentration, the shorter time required to be in contact. We must be careful during this process so that we don’t accidentally kill or destroyed the cells. 3. Callus Induction - After the sterilizing process, we then proceed to cutting the leaves into smaller pieces and putting them on the callus induction media. Again, do take note to always apply #aseptictechnique to prevent any contamination. In 2-3 days, we’ll come again to observe our plates, if there’s contamination, we will have to do some rescue and subculture. So yeah! We learned a lot! ✨ First Day FYP’s labwork - Media Preparation ✨ Hi everyone! Yesterday was the first day I started my final year project’s labwork at Agro-Biotechnology Institute (ABI), National Institutes of Biotechnology Malaysia. We prepared media for our experiments. It was a long process but i enjoyed every second of it. So, for my experiment, I need to regenerate shoot from callus culture. Theoretically, the auxin-cytokinin ratio for #shootregeneration is 1:4. I prepared 9 types of treatments to test out which one works the best to achieve my objective. In #plant #tissueculture, the most important aspect is sterilization, with that being said, we need to be EXTRA EXTRA careful and stay sanitized everytime handling the related materials and apparatus. In media preparation, some of the critical aspects are calculation of plant growth regulators’ concentration, pH of the media solution and again, sterilization (autoclave). Along the process, I realized that my lab skills are getting rusty but after a while, I got the hang of it and doing them more smoothly. All thanks to Dr Sharina and Encik Nadz for the demonstration and guiding us, and also Dr Radziah and Dr Hafiz for teaching us hands-on tissue culture techniques during our second year.
