A new study conducted by scientists Niranjan Nagarajan, Aarthi Ravikrishnan from A*STAR Genome Institute of Singapore (A*STAR GIS), in collaboration with researchers from Singapore Immunology Network, A*STAR Institute for Human Development and Potential (A*STAR IHDP) and the National University of Singapore has revealed that gut bacteria may play a key role in aging, and that specific microbes could be targeted to promote healthy aging. As part of this effort to understand the aging process and identify key lifestyle and dietary factors to encourage healthy aging, the team conducted the first large-scale study on over 200 octogenarians (people aged 80 - 89) in Singapore. Their investigation revealed how the gut microbiome changes with age, highlighting key microbial species associated with aging, and their potential effects on health. FISHing for answers in gene clusters Researchers A*STAR Genome Institute of Singapore (A*STAR GIS) devised a new streamlined method for analysing thousands of genes at once on a tissue-wide scale, offering a more accessible tool to study and diagnose diseases at a cellular level. “Spatial transcriptomics gives us both an unbiased overview of the biological pathways that underlie disease, and a detailed look at specific genes of interest,” said Principal Scientist Kok Hao Chen and Research Fellow Nigel Chou from A*STAR Genome Institute of Singapore (A*STAR GIS). However, the cost and time required for spatial transcriptomics currently limit its wider use in clinics and labs; imaging 20,000 genes with the latest multiplexed fluorescence in situ hybridisation (FISH) tool involves advanced equipment and weeks of data collection and analysis. To address those barriers, Chen, Chou and colleagues at GIS developed a streamlined form of FISH by focusing on essential aspects of disease analyses—such as identifying cell types and tissue patterns—without needing the high-resolution view of single RNA molecules. With RNA sequencing methods like nanopore sequencing, repetitive molecules may overshadow rare but important RNA types 🔍 As Yue Wan, Principal Investigator at the A*STAR Genome Institute of Singapore (A*STAR GIS), explained: “While a cell may express 5,000 to 15,000 genes at any time, roughly half of the resulting RNA molecules come from just 100 highly active genes, which usually aren’t research targets.” 💡 Wan and GIS colleagues collaborated with Oxford Nanopore Sequencing to test how adaptive sampling methods can filter out common RNA sequences and amplify rarer ones during RNA studies. When used on the transcriptome of 𝘾𝙖𝙣𝙙𝙞𝙙𝙖 𝙖𝙡𝙗𝙞𝙘𝙖𝙣𝙨, the team detected 26 novel RNA transcripts in the skin yeast, in addition to recording increased reads of target transcripts by 11 to 13.5%.
