    🔔 GATA3, a potential biomarker in breast cancer 🔔 🔬 Breast cancer commonly arises in luminal cells of the mammary gland expressing the estrogen receptor (ER). Patients with ER-positive breast cancer receiving endocrine therapy have initially a good prognosis, but studies with long-term follow-up have shown that there is a continuous risk of late relapse of the disease for these patients. Recent findings suggest that among ER-positive breast tumors, high nuclear and mRNA GATA3 expression, was seen in 84% and 41% of the cases, respectively. GATA3 expression is associated with ER-positive breast cancer and particularly with the Luminal A subtype.     🔔 Working on neurodegeneration? Calcium-binding proteins, calbindin and calretinin, are two important but less-explored member in this context. 🔬 Calcium, in addition to its essential role as a mediator of intracellular signaling, also serves as a key juncture in the process of neurodegeneration. Multiple injury pathways converge to induce an excessive rise in intracellular calcium levels which in turn activate a cascade of proteolytic enzymes, such as calpains and caspases, resulting in the onset of apoptosis. Maintenance of calcium homeostasis within neurons is essential to their well-being. Calbindin and calretinin are abundantly expressed through-out the central nervous system (CNS), and have been extensively studied due to their varying distributions, thus serving as markers of discrete neuronal subpopulations.     💡An important finding in Cardiometabolic Disease research... 🔎 A promising approach to improve cardiometabolic diseases (CMDs) is cold-induced thermogenesis (CIT), which increases resting metabolic rate and energy expenditure and reduces body weight, adipocyte size, and plasma glucose concentrations. In addition, CIT affects lipid metabolism by promoting the lipolytic clearance of triglyceride (TG)-rich VLDL by brown adipose tissue (BAT), leading to a reduction in circulating TG concentrations. 🔬 In a recent publication, Amor et al. have reported that fibrinogen alpha chain and fibronectin 1 are two important mediators involved in crosstalk between BAT and liver during CIT (Cardiovasc Diabetol. 2024; 23: 298).     🔔 OrgaPlexing: a novel approach to study cellular organelle coordination 🔔 🔬 Organelle communication is critical to connect metabolic pathways, whose subprocesses are segregated in different organelles across the cell. Zimmermann et al. have recently established OrgaPlexing, a multi-spectral organelle imaging approach that allows simultaneous visualization and behavioral analysis of up to six organelles, to examine the spatiotemporal coordination of organelle dynamics in primary macrophages (Nat Cell Biol. 2024; 26(8): 1261–1273). The approach provides a new experimental strategy to identify organelle-metabolic signalling hubs advancing our comprehension of organelle physiology and metabolism in both health and disease.
