    🔬 The intimate relationship between the epithelium and the immune system is essential for maintaining tissue #homeostasis. When this delicate balance is disrupted, it can lead to #autoimmunediseases and #cancer. While stem cell-derived #organoids have become powerful models for studying epithelial function, they fall short of capturing the complete organ-level processes due to the absence of tissue-resident immune cells. 🧫 Introducing human intestinal immuno-organoids (IIOs)! These groundbreaking models are formed through the self-organisation of epithelial organoids and autologous tissue-resident memory T (TRM) cells. A portion of these TRM cells integrate within the epithelium, continuously surveying the barrier. This interaction is driven by TRM cell-enriched transcriptomic programs that govern cell motility and adhesion, offering a deeper understanding of immune surveillance at the epithelial level. 🧬 By combining IIOs with single-cell transcriptomics, researchers (Timothy Recaldin et al.) investigated the complex dynamics of intestinal #inflammation triggered by cancer-targeting biologics in patients. The study uncovered that inflammation led to the emergence of an activated CD8+ T cell population, acquiring intraepithelial and cytotoxic features over time. This process was preceded by a T helper-1-like CD4+ population that initially produced cytokines and later became cytotoxic. 💡 The versatility of IIOs, coupled with their ability to mimic real immune interactions, allowed researchers to identify the Rho pathway as a potential target for mitigating immunotherapy-associated intestinal inflammation.     🧬 Unlocking Safe #Therapeutics: Gene Silencing with #Epigenetic Precision and Zinc Finger Technology 🧬 🔒 Gene silencing without the need for gene editing is paving the way for safer therapeutic applications. Researchers have developed an innovative strategy to repress multiple genes simultaneously using #ZincFinger proteins fused to Krüppel-Associated Box (KRAB) repression domains (ZF-Rs). This method offers a powerful tool for targeted gene modulation without altering the DNA sequence. 🦠 By optimising a lentiviral system for delivering ZF-Rs into hematopoietic cells, the authors (Davide Monteferrario, PhD et al.) from Sangamo Therapeutics, Inc., demonstrated that it's possible to multiplex up to three ZF-Rs, or even combine two ZF-Rs with a chimeric antigen receptor (#CAR). Impressively, the expression of these ZF-Rs had no negative impact on the integrity or functionality of the transduced cells, making this approach highly viable for therapeutic use. 📊 In both in vitro and in vivo experiments, ZF-R-expressing #Tcells achieved highly efficient gene repression over an extended period (up to 10 weeks). This repression was also linked to significant epigenetic remodeling, suggesting that ZF-Rs can effectively and sustainably modulate #gene expression without permanent genetic changes. 💡 To ensure #clinical safety, the researchers also developed methods to improve ZF-R #specificity, demonstrating a path toward the creation of ZF-Rs with a strong safety profile. Once misunderstood as a vestigial organ, our understanding of its complex functions has dramatically evolved since the groundbreaking work of Jacques Miller in 1961. The thymus is now recognized as essential for the development of #Tcells, with early T cell progenitors (ETPs) undergoing a series of intricate developmental steps within its structure. 🧪 Within the thymus, #thymocytes migrate from the outer cortex, where they undergo rapid proliferation and #PositiveSelection, to the inner medulla for #NegativeSelection. This process ensures that only T cells with appropriately regulated responses to self-antigens mature and enter circulation, while self-reactive T cells are eliminated or differentiated into regulatory T cells (#Tregs) to maintain immune tolerance. 🧩 Although these fundamental concepts have been established for decades, recent advancements in #technology have revolutionized our understanding of the thymus’s structure and function. Traditional methods like #histology, #immunohistochemistry, and #flowcytometry laid the groundwork, but they had limitations in #resolution and the #identification of diverse cell populations, especially within the stromal compartments. 🔬 Cutting-edge imaging and transcriptomics techniques, including high-resolution 3D imaging and single-cell #RNA sequencing, have provided unprecedented insights into the thymic #microenvironment.
