What are the differences between brightfield and fluorescence for cell counting / viability? Brightfield measurements using Trypan Blue are excellent for rapid reporting of cell counts and viability for cultured cell lines. However, the dye is subject to limitations in its ability to distinguish cells from debris. Dual fluorescence measurements using AO/PI enable the specific identification of live and dead cells in the presence of large numbers of non-nucleated cells and cellular debris. This removes the subjectivity associated with colorimetric dyes and improves the overall accuracy of results. Automated counting of SF9 insect cells 🐛 Insect cells such as the SF9 cell line isolated from Spodoptera frugiperda are commonly used in the process of recombinant protein production using baculovirus. This method of protein production requires accurate knowledge of SF9 cell counts and viability to qualify successful transfection and healthy cells producing protein. In this technical note, DeNovix scientists demonstrate how the CellDrop Automated Cell Counter accurately measures SF9 cells. Our application team tested two different viability dyes (Trypan Blue and AO/PI), and the CellDrop provided rapid and accurate measurements with both.
