Congratulations! It's exciting to see our SmartSPIM light sheet microscope featured in this groundbreaking study on enhancer AAVs targeting spinal motor neurons.
I'm proud to share our preprint on enhancer AAVs for selective targeting of spinal motor neurons and descending motor pathways. We generated rich 10X multiome datasets from the spinal cord of non-human primate and mouse, and used these to discover functional enhancers. We generated AAVs targeting the broad spinal motor neuron population or motor neuron subtypes. We demonstrate enhancers can be combined into a single vector to target both corticospinal tract and spinal motor neurons simultaneously. We extensively characterize the enhancer AAVs across in mouse, rat and NHP following diverse routes of administration and show stunning lightsheet data (LifeCanvas Technologies Inc.) and mFISH data (ACD Bio). Congratulations to the major contributors Emily Kussick, Nelson Johansen, Naz Taskin, Jonathan T. Ting and thank you to the many more team members that contributed to the study. I can't wait to see the major discoveries these tools will enable and am excited about their potential therapeutic application for devastating disorders such as ALS. Excited to share this new research, were the authors, including Joselyn Soto, Ph.D. at UCLA, use ΔFosB mapping to look at changes in neural activity patterns in the striatum and cortical areas in a mouse model of obsessive-compulsive disorder and their reversal using astrocyte-specific interventions. To do this, the authors utilized our #SHIELD post-fixation and #Clear+ delipidation protocols to optically clear brain tissue and #SmartSPIM light sheet microscope to capture the detailed 3D datasets for ΔFosB analysis. 🔗 Read full article: https://lnkd.in/gzt82RYT

Spinal motor neurons in dazzling detail. ✨ Each green dot is a spinal motor neuron - crucial but rare cells making up just 1% of neurons in the spinal cord. In diseases like ALS, they are selectively damaged, making them of special interest to neuroscientists, including this video's creators Tanya Daigle and Emily Kussick. They and their teams have developed a new, engineered version of the rabies virus to target spinal motor neurons and make it easier for scientists to study them. In the future, they hope this same technology can be used to deliver therapeutics that can help people living with neurodegenerative diseases. #SciShots

Celebrate #FluorescenceFriday with this new publication! In this work, the authors, including Xiaowei Li and liqin xiong from Shanghai Jiao Tong University, successfully utilized fluorescent polymer dots (Pdots) to achieve high-resolution, 3D imaging of the meningeal vascular network and the eye, opening new doors for understanding the intricate vascular structures in these regions.
