When acquiring a Z stack in thick samples, its typical for image brightness to be reduced with increasing distance from the objective lens due to effects such as scattering. Our latest #Microscopy #AppNote details the Z Intensity Correction function in our NIS-Elements software, which can be programmed to maintain uniform brightness throughout a Z stack by adjusting laser power and gain. An exciting new method combines in situ sequencing of genomic DNA with #SuperResolution expansion #Microscopy #ExM and labeling of nuclear proteins as a tool for correlating nuclear abnormalities with differences in gene regulation. The authors use this approach to spatially correlate abnormalities in the nuclear lamina with aberrant euchromatin repression in cells obtained from an individual with Hutchinson-Gilford progeria syndrome.
