Summary:

Expansion Therapeutics is a private well-funded biotechnology company focusing on developing transformative oral medicines for devastating RNA-mediated diseases. Expansion is seeking a highly motivated stem-cell biologist to play a key role in advancing the cross-functional drug discovery & development efforts. This position will be responsible for leading all stem-cell based in vitro disease modeling and assay development efforts and will be based in Expansion’s Cambridge, MA location.

Accountabilities & Responsibilities

  • Establish core stem cell capabilities by culturing, expanding, and banking multiple pluripotent stem cell lines.
  • Differentiate healthy donor and patient derived iPSCs to cardiomyocytes and neurons, as applicable to disease areas of interest.
  • Establish QC criteria, reproducibility, and robust consistency to scale up and develop iPS-derived cells to support industry-standard molecular, biochemical, and image-based assays using qPCR, ELISA, and/or flow cytometry, and high-content microscopy/imaging technologies.
  • Perform non-viral delivery of DNA and RNA oligonucleotides (ASOs, siRNAs, Anti-sense probes) into iPS-derived or other mammalian primary/immortal cells.
  • Develop, optimize protocols, and support generation of standard operating procedures (SOPs).
  • Support upkeep and maintenance of laboratory equipment.
  • Maintain detailed, organized records of experimental data and analysis.
  • Interpret and present data regularly in both oral and written formats.
  • Collaborate with and train scientists and colleagues across programs.
  • Perform all duties in keeping with the Company’s core values, policies and all applicable regulations.

Requirements

Requirements: Proven Experience, Skills, and Education:

  • PhD/Masters’ degree in a relevant biological field with hands-on 3-5-years of industry experience of iPS cell-based assay development and implementation.
  • Proven expertise in FISH and other immunofluorescence based cellular assays employing iPS-differentiated cells, confocal-microscopic imaging, and image analysis is a must.
  • Extensive experience in iPSC differentiation to cardiomyocyte lineages is highly valued.
  • Additional experience in iPSC differentiation to skeletal muscle and/or neuronal lineages will be appreciated.
  • Knowledge of RNA-mediated diseases and nucleotide-repeat-expansion disorders will be highly valued.
