Our preprint on modelling chromosome instability and structural variation in now on bioRxiv! Work led by BingXin L. https://lnkd.in/eiYuRcSd We developed a cell-cycle dependent break/repair/replication computational model and fit it to single-cell genomic data. The model captures the essentials of what happens in G1, S, G2 and M when double strand breaks (DSBs) occur. When there are no cellular checkpoints DSBs are allowed to propagate into S and G2. When these are joined via NHEJ they causing (sometimes massive) rearrangement. What's really cool about this model is that is unifies various phenomena including chromothripsis, ecDNA, breakage fusion bridge cycles and seismic amplification. And shows that NHEJ is sufficient to give rise to all the different types of rearrangements. Please get in touch if you are interested in this work, we are open to collaborations!
